摘 要:目的探讨艾灸足三里穴对功能性消化不良模型大鼠肠神经胶质细胞的影响。方法将50只大鼠随机分为正常组(10只)及造模组(40只),将造模成功的30只大鼠再随机分为模型组、艾灸组及束缚组,每组10只。造模结束后,艾灸组大鼠用自制鼠衣束缚后艾灸足三里穴,束缚组只进行束缚。干预结束后取大鼠胃窦组织,用HE染色观察大鼠胃窦组织病理形态变化,免疫荧光染色观察S100β、GFAP蛋白表达量,PCR观察大鼠胃窦组织s100β、GFAP mRNA水平。结果大鼠腹部解剖显示:各组大鼠胃组织未发现器质性改变。s100β、GFAP蛋白免疫荧光染色阳性为红色荧光,半定量分析结果显示,模型组胃窦组织s100β、GFAP蛋白相对表达量显著高于正常组(P<0.01);与模型组比较,艾灸组大鼠胃窦组织中s100β、GFAP蛋白相对表达量明显降低(P<0.01);束缚组与模型组之间没有统计学差异;与束缚组相比,艾灸组大鼠胃窦组织中s100β、GFAP蛋白相对表达量明显降低(P<0.01)。与正常组相比,模型组大鼠胃窦组织S100βmRNA、GFAP mRNA表达明显升高(P<0.01);与模型组相比,艾灸组大鼠胃窦组织S100βmRNA、GFAP mRNA表达明显降低(P<0.01),而束缚组与模型组差异无统计学意义;与束缚组相比,艾灸组S100βmRNA、GFAP mRNA表达明显下降(均P<0.01)。结论在功能性消化不良模型大鼠中,存在着EGCs的异常增生及活化,而艾灸足三里穴可以通过调节EGCs的异常增生及活化来改善功能消化不良相关症状。Objective:To investigate the effects of moxibustion at Zusanli point on enteric glia cells in rats of functional dyspepsia(FD).Methods:50 rats were randomly divided into the normal group(10 rats)and the modeling group(40 rats).The modeling group were modeled,and 30 rats successfully modeled were randomly divided into the model group(MG),moxibustion group(MBG)and restraint group(RG)with 10 rats in each group.The rats in the MBG were restrained and suspended,and receive the moxibustion at Zusanli point.RG were simply restrained.At the end of the intervention,the gastric antrum tissues of rats were taken.HE staining was used to observe the pathological changes of gastric antrum;the expression of s100βand GFAP protein was observed by immunofluorescence staining,and the mRNA levels of s100βand GFAP were observed by PCR.Results:Abdominal anatomy of the rats showed no organic changes in the gastric tissues of the rats in each group.s100βand GFAP immunofluorescence staining showed that the relative expression of s100βand GFAP in MG was significantly higher than that in NG;compared with MG,that in MBG significantly decreased.There was no statistical difference between RG and MG;compared with RG,that in MBG was significantly reduced.Compared with NG,the mRNA expression of S100βand GFAP in MG was significantly increased;compared with MG,that in MBG was significantly decreased,and there was no significant difference between the RG and the MG;compared with RG,that in MBG was decreased significantly(P<0.01).Conclusion:In FD model rats,there is abnormal proliferation and activation of EGCs,and the moxibustion at Zusanli point can improve the symptoms of functional dyspepsia by regulating the abnormal proliferation and activation of EGCs.
