川芎苦参不同比例配伍对活性成分和抗氧化活性的影响

摘　　要：目的观察川芎与苦参不同比例配伍后对川芎嗪和苦参碱成分的溶出和抗氧化活性的影响,探讨川芎、苦参中药配伍的意义。方法实验设川芎与苦参比例设为9组,分别为3∶1、2.5∶1、2∶1、1.5∶1、1∶1、1∶1.5、1∶2、1∶2.5、1∶3。分别检测提取液中川芎苦参活性成分川芎嗪、苦参碱的含量,并DPPH法测定各提取液的抗氧化活性。结果川芎和苦参不同比例配伍对活性成分川芎嗪、苦参碱含量具有明显差异,其中,川芎、苦参配伍比例为3∶1时川芎嗪含量最高,2∶1时苦参碱含量最高。不同比例配伍对抗氧化活性有显著影响,其中,川芎、苦参配伍比例为1∶2时抗氧化活性最强,1.5∶1时抗氧化活性最弱。结论川芎和苦参在不同比例配伍下,有效成分川芎嗪和苦参碱含量溶出有明显的差异,体现出不同的抗氧化活性,可为临床辨证治疗不同证型合理选药及优化设计剂量组合提供依据。Objective:To observe the effect of different proportions of Ligusticum wallichii and Sophora flavescens on the dissolution and antioxidant activity of Ligustrazine and matrine,and to explore their significance of compatibility.Methods:Nine experimental groups were set up in the ratio of 3∶1,2.5∶1,2∶1,1.5∶1,1∶1,1∶1.5,1∶2,1∶2.5,1∶3 of Ligusticum wallichii and Sophora flavescens respectively.The contents of ligustrazine and matrine were detected,and their antioxidant activity was determined by DPPH.Results:The content of ligustrazine and matrine in different proportion of Ligusticum chuanxiong and Sophora flavescens were significantly different.The content of ligustrazine was the highest when the ratio of Ligusticum wallichii and Sophora flavescens was 3∶1,and the content of matrine was the highest when the ratio was 2∶1.The antioxidant activity was significantly affected by different proportion.Among them,when the ratio was 1∶2,the antioxidant activity was the strongest,and the antioxidant activity was the weakest at the ratio of 1.5∶1.Conclusions:When Ligusticum wallichii and Sophora flavescens are combined in different proportion,there are obvious differences in the content of Ligustrazine and Matrine,and they have different antioxidant activities,which provide the basis for the rational selection of drugs and the optimal design of dosage combination for different syndrome types in clinical syndrome differentiation and treatment.
