养殖中华乌塘鳢致病性弧菌的分离鉴定

摘　　要：2017年8月,浙江台州某养殖场的中华乌塘鳗出现皮肤溃疡病,症状主要表现为体表病灶部位出血、肌肉溃烂等。解剖发现病鱼肝脏病变呈土黄色,脾脏充血肿大,肠道后端有黄色粘液。组织病理切片显示患病中华乌塘鳗肝脏和脾脏均出现严重的变性坏死。从病鱼体表溃疡部位及内脏分离到1株优势菌并命名为TZ-01,经人工回归感染实验证明TZ-01即为引起本次养殖中华乌塘鳗皮肤溃疡病的病原菌。该菌株在硫代硫酸盐柠檬酸盐胆盐蔗糖琼脂培养基(Thiosulfate citrate bile salts sucrose agar culture medium,TCBS)上形成圆形菌落,微隆起,呈蓝色或蓝绿色,革兰氏染色呈阴性,短杆状。结合API-20E快速鉴定、16S rDNA同源性分析等方面的结果,鉴定TZ-01菌株为副溶血弧菌。药敏试验证实,TZ-01菌株对头抱曲松、头抱他曉、米诺环素、多西环素、四环素等多种抗生素敏感。An outbreak of ulcer disease of cultured Chinese black sleeper Bostrychus sinensis with the symptoms such as focal hemorrhage and muscle ulceration,has occurred in a farm in Taizhou,Zhejiang Province in August 2017.Anatomical observation showed severe degeneration and necrosis in both liver and spleen.A dominant strain of bacterial pathogen(named TZ-01)was isolated from ulceration site and internal organs of the diseased fish.Artificial infection proved that the isolated bacterium was the pathogen causing ulcer disease of cultured Chinese black sleeper.The strain,which was Gram-negative and short rod,could grow in the TCBS culture medium and form protruding,blue colonies.Combined with results from API-20E identification system and 16S rDNA sequence analyses,TZ-01 was identified as Vibrio parahaemolyticus.The drug sensitivity test showed that TZ-01 was sensitive to several antibiotics,including ceftriaxone,ceftazidime,minocycline,doxycycline and tetracycline.
