摘 要:目的通过建立大鼠慢性软组织损伤实验动物模型,初步探讨针刀对慢性软组织损伤的治疗机制。方法将2月龄SPF级SD雄性大鼠总计80只随机分为对照组(20只),模型组(20只),电刀组(20只),针刀组(20只)。模型组采用Kami机械冲击挫伤的造模方法,造模成功后采用电动针刀及针刀2种不同口径的针刀进行干预治疗,分别于第6天、12天取材进行组织形态学观察,采用蛋白质免疫印迹(Western-blot)检测不同组别大鼠软组织局部组织中转化生长因子-β(TGF-β)及血管内皮生长因子(VEGF)表达量的差异。结果组织形态学HE染色观察发现针刀组大鼠损伤肌肉组织修复与其他组比较有差异,蛋白质免疫印迹(Western-blot)检测结果显示第6天时针刀组TGF-β及VEGF蛋白表达量明显高于其他组,差异均有统计学意义(P<0.05),第12天时各组TGF-β及VEGF蛋白表达量均有所下降,但是针刀组2种生长因子的表达量处于相对较高水平,VEGF蛋白表达量明显高于其他组,而TGF-β蛋白表达量略低于其他组。结论针刀对治疗慢性软组织损伤有一定促进作用,能够提高大鼠损伤组织中TGF-β及VEGF蛋白表达量,促进组织恢复。Objective To establish a laboratory model of chronic soft tissue injury in rats,and to explore the therapeutic mechanism of needle knife on chronic soft tissue injury.Methods A total of 80 male SPF SD male rats were randomly divided into a blank control group(20 rats),a chronic soft tissue injury model group(20 rats),a electric needle knife treatment groupⅠ(20 rats)and ordinary needle knife treatment groupⅡ(20 rats).The model group used Kami mechanical impact contusion modeling method.After successful modeling,the electric needle knife and ordinary needle knife were used to treat the lesions.The histomorphological observation was carried out on the 6th and 12th days respectively.Western blotting was used to detect the differences of TGF-βand VEGF expression in the soft tissue of the different groups of rats.Results HE staining showed that there were differences in muscle tissue repair in rats treated with ordinary needle knife compared with other groups.On the 6th day,the expression of TGF-βand VEGF protein in the ordinary needle knife group was significantly higher than that in the other groups(P<0.05).On the 12th day,the expression of TGF-βand VEGF protein decreased.However,the expressions of TGF-βand VEGF in the ordinary needle treatment group II was significantly higher than that in the other groups,and the expression of VEGF was significantly higher than that in the other groups,while the expression of TGF-βwas slightly lower than that in the other groups.Conclusion Needle knife can be used to treat chronic soft tissue injury through the mechanism of“needle knife treatment-microcirculation-platelet-growth factor-tissue repair”to complete the repair of damaged soft tissue.
