摘 要:目的研究理冲汤对子宫肌瘤模型小鼠Th1/Th2平衡漂移的调控作用。方法将24只CB-17 Scid雌性小鼠随机分为4组,正常组、模型组、理冲汤组和阳性药物组,采用异种移植法制备小鼠子宫肌瘤模型,药物干预28天后,采用HE染色来观察各组小鼠子宫平滑肌结构,采用MSD超敏多因子电化学发光法检测小鼠血清中Th1细胞分泌的IL-2、IFN-γ和Th2细胞分泌的IL-4、IL-5、IL-10的含量水平。结果HE染色结果显示模型组小鼠子宫平滑肌组织及腺体较之于正常组有明显增生,细胞排列紊乱,而理冲汤组和阳性药物组小鼠子宫平滑肌组织基本正常。MSD检测结果显示与正常组比较,模型组小鼠血清中的IL-2、IFN-γ的浓度显著降低(P<0.01),而IL-4、IL-5、IL-10的浓度显著升高(P<0.05,P<0.01);阳性药物组的IL-4、IL-5、IL-10的浓度也显著升高(P<0.05,P<0.01),理冲汤组仅IL-5的浓度较之于正常组显著升高(P<0.01)。与模型组比较,理冲汤组小鼠血清中的IL-2、IFN-γ均显著升高(P<0.05,P<0.01),IL-4、IL-5、IL-10的浓度均显著降低(P<0.05,P<0.01);而阳性药物组中的IL-2、IFN-γ均显著升高(P<0.05,P<0.01),但IL-4、IL-5、IL-10的浓度变化比较,差异无统计学意义(P>0.05)。结论理冲汤通过调控Th1、Th2细胞分泌因子的含量水平,影响免疫逃逸,进而发挥抑制子宫肌瘤的作用。Objective To study the regulation effect of Lichong Decoction on Th1/Th2 drift of balance in mice model with uterine fibroids.Methods Twenty-four female CB-17Scid mice were randomly divided into four groups:normal group,model group,Lichong Decoction group and positive drug group.Mouse model of uterine fibroids was prepared by heterograft method.Drug intervention after 28 days,the structure of uterine smooth muscle was observed by HE staining and the levels of cytokines IL-2,IFN-γsecreted by Th1 cells and IL-4,IL-5 and IL-10 secreted by Th2 cells in serum of mice were detected by MSD hypersensitive multi-factor electrochemiluminescence method.Results The results of HE staining showed that the uterine smooth muscle tissue and glands in the model group had obvious hyperplasia and the cells were disordered compared with the normal group,but the uterine smooth muscle tissue of Lichong Decoction group and positive drug group was normal,so we built the model successfully.MSD results showed that compared with the normal group,the concentrations of IL-2 and IFN-γin the model group were significantly decreased(P<0.01),but the serum concentrations of IL-4,IL-5 and IL-10 were significantly increased(P<0.05,P<0.01),and the concentrations of IL-4,IL-5 and IL-10 were also significantly increased in the positive drug group(P<0.05,P<0.01),and only the concentration of IL-5 in the Lichong Decoction group was significantly higher than that in the normal group(P<0.01).Compared with the model group,the serum concentrations of IL-2 and IFN-γwere significantly increased in the Lichong Decoction group(P<0.05,P<0.01),and the concentrations of IL-4,IL-5 and IL-10 in the serum of mice were significantly decreased(P<0.05,P<0.01).However,in the positive drug group,only IL-2 and IFN-γwere significantly increased(P<0.05,P<0.01),and the changes in IL-4,IL-5 and IL-10 concentrations were not statistically significant(P>0.05).Conclusion Lichong Decoction affects immune escape by regulating the level of secreted factors of Th1 and Th2 cells,th
