摘  要:目的建立痔炎消颗粒的HPLC指纹图谱。方法采用HPLC法,色谱柱为Agilent ZORBAX Eclipse XDB-C18(4.6 mm×250 mm,5μm);以乙腈-0.1%磷酸水溶液为流动相进行梯度洗脱;体积流量:1.0 mL·min^-1;柱温:30℃;进样量:20μL;检测波长:230 nm。采用指纹图谱相似度评价软件,再结合聚类分析(CA)、主成分分析(PCA)及正交偏最小二乘判别分析(OPLS-DA)对10批痔炎消颗粒指纹图谱进行评价。结果 10批痔炎消颗粒HPLC指纹图谱中有15个共有峰,并鉴定出了3个成分(绿原酸、芦丁、柚皮苷),指纹图谱相似度均在0.900以上,各批样品一致性较好。通过CA、PCA、OPLS-DA分析,10批样品按不同生产厂家聚为3组,与样品来源相符,并筛选出包括柚皮苷在内的3个对分组贡献最大的组分。结论所建立的方法简便、重复性好,可为痔炎消颗粒的整体质量评价提供基础数据。Objective To establish a HPLC chromatographic fingerprint of Zhiyanxiao granules.Methods The chromatographic fingerprint of Zhiyanxiao granules was developed with HPLC.The separation was carried out on an Agilent ZORBAX Eclipse XDB-C18 column(4.6 mm×250 mm,5 μm) with the mobile phase consisting of acetonitrile and 0.1% phosphoric acid at a flow rate of 1.0 mL·min^-1.The column temperature was 30℃,the sample volume was 20 μL,and the detection wavelength was 230 nm.Similarity evaluation combined with cluster analysis(CA),principal components analysis(PCA) and orthogonal partial least squares discriminant analysis(OPLS-DA)were used to evaluate the HPLC chromatographic fingerprint of 10 batches of Zhiyanxiao granules.Results The common mode of the chromatographic fingerprint was set up with 15 common peaks,and 3 of them were identified,including chlorogenic acid,rutin and naringin.The similarity degrees of 10 batches of samples were more than0.900,and showed that all the samples were of good consistency.Ten batches of samples were classified into 3 clusters according to different manufacturers,which were consistent with the source of samples,and 3 components including naringin contributed most to the classification were obtained.Conclusion The method is simple and reproducible,and provides essential data for the overall quality evaluation of Zhiyanxiao granules.
