摘 要:目的:通过地龙降压胶囊对SHR大鼠凝血相关的血小板miRNAs表达的影响,探讨地龙降压胶囊的降压机制。方法:SHR大鼠分为5组:正常对照组、肝素阳性对照组、地龙降压胶囊低、中、高剂量组。药物高、中、低组分别灌胃地龙降压胶囊100 mg/kg、50 mg/kg、25 mg/kg。15 d后取血,实时荧光定量PCR(RT-PCR)方法检测5种miRNAs的表达水平。结果:与正常对照组相比,地龙降压胶囊干预组与血小板活化相关的miRNA-223、miRNA-96和聚集相关的miRNA-130a的表达量明显改变,而与血小板生成有关的miRNA-155、miRNA34a-5p的表达量无明显变化。结论:地龙降压胶囊可能主要通过影响血小板活化与聚集相关的miRNAs发挥抗凝血降压作用。Objective To explore the hypotensive mechanism of Dilong antihypertension Capsule, based on the investigations of the expressions of miRNAs coagulation-related platelets after the administration of Dilong antihypertension Capsule. Methods SHR rats were randomly divided into 5 groups: normal control group, heparin-positive control group, low-dose group of Dilong antihypertension Capsule, medium-dose group of Dilong antihypertension Capsule, high-dose group of Dilong antihypertension Capsule. The high, medium and low-dose groups were respectively given Dilong antihypertension Capsule 100 mg/kg, 50 mg/kg and 25 mg/kg. Bloods were collected 15 days after administration, and the expressions of five miRNAs were detected by real-time fluorescence quantitative PCR(RT-PCR). Results Compared with the control group, the expressions of platelet activation-related miRNA-223, miRNA-96 and conglomeration-related miRNA-130 a in Dilong antihypertension Capsule-treated groups changed significantly, while there was no obvious change in the expressions of miRNA-155 and miRNA34 a-5 p related to thrombocytopoiesis. Conclusion Dilong antihypertension Capsule may play an anticoagulant and hypotensive role mainly by affecting the miRNAs expressions of platelet activation and aggregation.
