摘  要:目的:探讨黄芩素对重症急性胰腺炎(SAP)大鼠肺组织损伤的影响及机制。方法:将60只SPF级SD大鼠分为假手术组、SAP组和SAP+黄芩素组,每组20只。SAP组和SPA+黄芩素组采用牛磺胆酸钠法建立SAP模型后5 min尾静脉注射10 g/L黄岑素2 mL/kg,于术后12、24 h(每个时间点每组10只)采用酶联免疫吸附法、全自动生化分析仪分别检测血清IL-6、TNF-α及淀粉酶含量,酶联免疫吸附法检测肺组织中MDA含量和SOD活性,TUNEL法检测肺组织细胞凋亡指数,Western blot法检测肺组织中Bcl-2、Bax和p-p38MAPK蛋白的表达。结果:与假手术组比较,SAP组大鼠术后12、24 h血清淀粉酶、IL-6和TNF-α水平及肺组织MDA含量、细胞凋亡指数升高,SOD活性降低,肺组织中Bax和p-p38MAPK蛋白表达升高,Bcl-2蛋白表达降低(P<0.05)。与SAP组比较,SAP+黄芩素组大鼠术后12、24 h血清淀粉酶和IL-6、TNF-α水平及肺组织MDA含量、细胞凋亡指数降低,SOD活性升高,肺组织中Bax和p-p38MAPK蛋白表达降低,Bcl-2蛋白表达升高(P<0.05)。结论:黄芩素可减弱SAP大鼠炎症反应、氧化应激、肺组织细胞凋亡,从而对肺组织起保护作用,机制可能与抑制p38MAPK信号通路有关。Aim:To explore the effects and mechanism of baicalein on lung tissue injury in rats with severe acute pancreatitis(SAP).Methods:Sixty SPF SD rats were randomly allocated into sham operation group,SAP group and SAP+baicalein group,with 20 rats in each group.SAP model was established by sodium taurocholate method.Serum IL-6,TNF-αand amylase were detected by ELISA and automatic biochemical analyzer at 12 and 24 hours after operation.MDA content and SOD activity in lung tissue were detected by ELISA.The cell apoptosis index of lung tissue was detected by TUNEL method.Western blot was used to detect the expressions of Bcl-2,Bax and p-p38MAPK protein in lung tissue.Results:Compared with the sham operation group,the levels of amylase,IL-6 and TNF-αin serum and MDA content and cell apoptosis index in lung tissue of SAP group were increased significantly at 12 and 24 hours after operation,SOD activity was decreased,the expressions of Bax and p-p38MAPK protein in lung tissue were increased,and the expression of Bcl-2 protein was decreased(P<0.05).Compared with SAP group,the levels of amylase,IL-6 and TNF-αin serum and MDA content and cell apoptosis index in lung tissue of SAP+baicalein group were decreased significantly at 12 and 24 hours after operation,SOD activity were increased,the expressions of Bax and p-p38MAPK protein in lung tissue was decreased,and the expression of Bcl-2 protein was increased(P<0.05).Conclusion:Baicalein could decrease the inflammatory reaction,oxidative stress and cell apoptosis of lung tissue in SAP rats,and has protective effects on lung tissue.The mechanism may be related to the inhibition of p38MAPK signaling pathway.
