摘  要:通过比较活体成像系统中不同玉米芽长、曝光时间及合并像素值的荧光素酶信号强弱,优化利用活体成像系统筛选转荧光素酶玉米植株的条件。结果表明,在玉米芽长3~8 cm、喷施底物反应2 min后曝光时间5 min、像素合并值binning 8×8的条件下,能够较好地检测到植株荧光素酶信号;进一步对获得的转基因玉米植株进行活体检测,确定了信噪比值大于2.0的植株为强表达阳性植株。研究结果可为转基因植株的后代筛选提供新方法,可以淘汰大量非转基因和表达较弱的植株或株系,有效提高筛选效率。The expression of luciferase gene in transgenic maize plants was detected by in vivo imaging system.The method was optimized through comparing different parameters such as the length of maize buds,the exposure time and the pixel combination value.The results showed that the luciferase signal could be better detected,when the maize bud was 3-8 cm,the exposure time was 5 min and the pixel combination valu was 8×8 binning,the reaction time was 2 min after spraying substrate.The strongly expressed transgenic plants were determined with the signal-to-noise ratio(SNR)more than 2.0.This study provided a new method for screening transgenic plants,which could eliminate a large number of non-transgenic and poorly expressed plants or lines so that it reduced the workload of screening transgenic plants,and improved the screening efficiency.
