摘 要:为探究禽网状内皮组织增殖病病毒(REV)感染对雏鸡胸腺、脾和法氏囊细胞凋亡及相关基因c-Myc表达、核质比值的影响,并阐述其相互关系。以1日龄SPF雏鸡为研究对象,应用分子生物学和免疫学技术结合病理学检测方法,通过对上述免疫器官c-Myc基因m RNA及其蛋白含量、c-Myc阳性细胞数、细胞凋亡率及核质比值的检测,较全面系统地研究了REV感染SPF雏鸡后第1、7、14、21、28和42天上述被检指标的动态变化。结果显示,REV感染雏鸡后,无论中枢免疫器官(胸腺、法氏囊)还是外周免疫器官(脾),其c-Myc基因mRNA表达和蛋白含量及阳性细胞数量均不同程度的高于对照雏鸡,其中21~28 d差异显著(P<0.05)或极显著(P<0.01);免疫器官细胞凋亡率在病毒感染后14~28 d极显著(P<0.01)高于对照雏鸡;免疫器官细胞核质比值不同程度的小于对照雏鸡,特别是病毒感染后21~28 d差异显著(P<0.05),以法氏囊细胞凋亡和核质比变化尤为明显;细胞凋亡率和核质比值变化具有较好的相关性(R^2=0.812,P<0.01)。表明REV感染所致免疫器官细胞凋亡与c-Myc基因过表达密切相关,核质比值减小程度可间接反映细胞凋亡程度。This research was conducted to study cell apoptosis,expression of c-Myc gene and nucleoplasm ratio(N/C) in immune organs of specific pathogen-free(SPF) chicks infected with reticuloendotheliosis virus(REV). In this article,1-day-old chicks were used, molecular biology,immunological techniques and pathological detection methods were applied to test the changes of c-Myc gene expression and protein content,number of c-Myc positive cells,apoptotic rate and N/C in immune organs at 1,7,14,21,28 and 42 days.The results showed that in REV infected chicks,the content of c-Myc and the number of c-Myc positive cells in immune organs were significantly higher than control chicks at 21-28 dpi.The apoptotic rate of cells in immune organs was significantly higher than that in control chicks at 14-28 dpi,while N/C was significantly lower than that in control chickens at 21-28 dpi.There was a good correlation between cells apoptotic rate and N/C which suggested that the cells apoptosis in immune organs induced by REV infection is closely related to the overexpression of c-Myc gene,and the reduction of N/C can directly reflect the progress of apoptosis.
