摘  要:为了探讨高温条件对猪睾丸支持细胞(stertoli cells)免疫豁免相关基因的影响,笔者分离培养猪原代sertoli细胞,利用差速贴壁法进行纯化,经sertoli细胞标记基因核酸与蛋白的检测鉴定,分离的原代sertoli细胞倍增时间为27.5 h。对原代sertoli细胞分别进行40℃与43℃热应激处理,并给予不同的热处理时间(0.25、1、6 h)与热应激恢复时间(0.5、1、2 h)处理,检测细胞内及上清中FasL与TGF-β基因的表达与蛋白的分泌差异。结果表明,热应激处理会使原代sertoli细胞FasL与TGF-β基因的表达量下降,但蛋白的分泌均上调,不同的热处理温度与时间并未有显著差异。热应激后复温处理对细胞内基因的表达有恢复作用,但是对细胞上清蛋白的分泌并没有显著影响,恢复时间在0.25 h^2 h内没有显著差异。To investigate the effects of high temperature on the immune privilege related gene of swine sertoli cells,primary sertoli cells were isolated from swine testis,and purified using the method of differential attachment,and identified using detecting nucleic acid and protein marker genes of sertoli cells. The double time of primary stertoli cell is 27.5 h.Primary sertoli cells were treated for heat stress at 40 ℃ and 43 ℃ for 0.25 h,1 h and 6 h respectively,and cells were recovery cultured at 37 ℃ for 0.5 h,1 h,2 h respectively after heat stress treatment.The Fas L and TGF-β genes’ expression and protein secretion were detected.The results showed that Fas L and TGF-β genes expression of intracellular were decreased while proteins secretions of cellular supernatant were increased under heat stress treatment.Gene expression and protein secretion did not have significant differences between 40 ℃ and 43 ℃.The FasL and TGF-β genes expression of intracellular were recovered by heat stress and recovery treatment,but the secretion of protein in the supernatant had no significant differences within 0.25 h-2 h.
