摘  要:目的系统分析脂多糖(LPS)感染过程中,人参二醇对心脏微血管内皮细胞(CMECs)基因表达的影响。方法通过分离大鼠原代CMECs,将细胞分为对照组和实验组,对照组用LPS单独刺激,实验组用LPS和人参二醇联合刺激,之后利用转录组测序技术整体分析差异表达的基因,以及相关信号通路,并通过定量PCR对部分差异表达基因进行验证。结果人参二醇能显著抑制部分基因的表达,且主要富集于炎症信号相关通路、代谢相关通路和表观遗传调节等。结论人参二醇可能通过表观遗传调控的方式影响炎症相关基因的表达,进而调控CMECs的炎症反应。Objective To explore the effect of 20R-Panaxadiol on the expression of cardiovascular endothelial cells during lipopolysaccharide(LPS)infection.Methods Cardiac microvascular endothelial cells of mice were isolated and divided into control group and observation group,and then stimulated by LPS in control group or LPS combined with 20R-Panaxadiol in observation group.The differential expression of genes and related signal pathways were analyzed using integrated transcriptome sequencing technology.Quantitative PCR was used to verify some differentially expressed genes.Results 20R-Panaxadiol significantly inhibited the expression of some genes with mainly involved in inflammatory signaling pathways,metabolic pathways and epigenetic regulation.Conclusion 20R-Panaxadiol affects the expression of inflammatory related genes to regulate inflammatory response of cardiac microvascular endothelial cells through epigenetic regulation.
