摘  要:目的观察密蒙花颗粒剂对去势雄兔泪腺细胞Bax、Caspase-3、Fas和FasL表达的影响:方法30只新西兰长耳白兔随机平均分为5组,分别为:A组:空白组,不灌胃;B组:模型组,不灌胃;C组:密蒙花颗粒剂组,灌胃药物为密蒙花颗粒剂;D组:安慰剂组,灌胃药物为生理盐水;E组:睾酮组,在兔大腿肌肉处注射丙酸睾酮注射液,,除A组外,所有兔子在干预前的造模方式均为双侧睾丸及附睾切除术,并在造模前1天和造模后第30天测定SIT和BUT。干预30天后,将所有兔子处死,摘取泪腺组织进行伊红染色观察泪腺组织结构,免疫组化法检测各组泪腺组织中Bax、Caspase-3、Fas和FasL表达结果HE染色显示:密蒙花颗粒剂组兔子泪腺组织排列整齐,细胞结构清晰,有少量炎症细胞浸润和细胞凋亡,Bax sCaspase-3、Fas和FasL少量表达于细胞膜和细胞浆中免疫组化检测显示密蒙花颗粒剂组Bax、Caspase-3、Fas和FasL平均光密度均较模型组减少(P<0.01).结论密蒙花颗粒剂对去势雄兔泪腺细胞Bax、Caspase-3、Fas和FasL表达具有抑制作用;密蒙花颗粒剂通过抑制泪腺组织中Bax xCaspase-3、Fas和FasL的表达减轻泪腺组织的凋亡,达到治疗干眼的目的。Objective To observe the effects of Mimeng granules on the expression of Bax,Caspase-3,Fas and FasL in ovariec-tomized male rabbit lacrimal gland cells.Methods Thirty New Zealand long-eared white rabbits were randomly divided into 5 groups:group A:blank group,no gavage;group B:model group,no gavage;group C:Mimenghua granule group,gavage drug for the Mimenghua granules;Group D:placebo group,the saline drug was normal saline;Group E:testosterone group,testoster­one propionate injection was injected into the rabbit thigh muscle.Except for group A,all rabbits were treated with bilateral tes­ticular and epididymal resection before intervention,and SIT and BUT were measured 1 day before modeling and 30 days after modeling.After 30 days of intervention,all rabbits were sacrificed.Lacrimal gland tissue was harvested for eosin staining to ob­serve the structure of lacrimal gland.Immunohistochemistry was used to detect tlie expression of Bax,Caspase-3,Fas and FasL in lacrimal gland tissue.Results HE staining showed that the lacrimal gland of rabbits in the Mimenghua granule group was neatly arranged,the cell structure was clear,and there was a small amount of inflammatory cell infiltration and apoptosis.Bax,Caspase-3,Fas and FasL were expressed in the cell membrane and cytoplasm.Immunohistochemistry showed that the average optical densities of Bax,Caspase-3,Fas and FasL in the Mimenghua granule group were lower than those in the model group(P<0.01).Conclusion Mimenghua granules inhibited the expression of Bax,Caspase—3,Fas and FasL in ovariectomized rab­bit lacrimal gland cells;Mimenghua granules alleviated lacrimal gland tissue by inhibiting the expression of Bax,Caspase-3,Fas and FasL in lacrimal gland tissue The apoptosis is achieved for the purpose of treating dry eyes.
