扶芳藤提取物抗HIV-1活性初步实验研究

摘　　要：目的观察扶芳藤15%乙醇提取部位的抗HIV-1作用,为寻找新的有效治疗艾滋病药物提供依据方法以TZM-bl-HIV-1ⅢB系统和MT2-HIV-1ⅢB系统为实验模型,使用荧光素酶活性检测试剂检测病毒复制水平,ATP法检测不同浓度扶芳藤提取物作用下细胞的相对活力;通过酶标仪检测化学发光信号,计算相对病毒水平和相对细胞活力结果扶芳藤提取物在TZM-bl-HIV-1ⅢB系统和MT2-HIV-1ⅢB系统中,可在较大浓度范围对病毒感染产生抑制作用,而不对细胞造成较大毒性,因此该提取物为扶芳藤抗HIV-1中药有效部位结论扶芳藤15%乙醇提取部位,抗HIV-1效果明显,为今后深入研究其药用价值和临床意义提供了更为有力的理论基础.Objective To observe the anti - HIV -1 effect of 15% ethanol extraction site of the Euonymus Fortunei extract and provide basis for finding new effective drugs for the treatment of AIDS. Methods The TZM - bl — HIV - 1ⅢB system and the MT2 - HIV -1ⅢB system were used as experimental models. The level of viral replication was detected by using a luciferase ac­tivity assay reagent. The ATP method was used to measure the relative viability of cells under different concentrations of amygda- lin. The chemiluminescent signal was detected by a microplate reader to calculate relative viral levels and cell viability. Results The extract of Euonymus fortunei can inhibit the infection of the virus in a large concentration range without causing great toxicity to the cells , so this extract is the effective part. Conclusion The 15% ethanol extraction site of the Euonymus Fortunei has obvious anti - HIV - 1 effect, which provides a more powerful theoretical basis for further study of its medicinal value and clinical significance.
