养心汤对Hey诱导内皮损伤大鼠血管舒张功能的影响

摘　　要：目的观察养心汤对Hey诱导内皮损伤大鼠血管舒张功能的影响方法雄性SD大鼠40只,随机分为空白组、模型组、养心汤组、叶酸组,每组10只空白组给予普通饲料,其余4组给予含3%蛋氨酸饲料喂养12周,复制高同型半胱氨酸血症血管内皮损伤模型第13周全部大鼠给予常规普通饲料喂养同时进行干预,模型组和空白组大鼠给予常规等容积蒸饱水灌胃,养心汤组给予养心汤浸膏2.34g/(kg•d)灌胃,叶酸组给予叶酸1.8mg/(kg•d)灌胃,连续给药4周;第16周末,大鼠麻醉称重取材,ELISA法检测血清NO、ET-1水平;离体血管环检测大鼠胸主动脉血管舒张功能:结果与模型组比较,养心汤能够增加血清NO,降低ET-1含量;改善内皮依赖性血管舒张功能,结论养心汤能够降低ET-1、上调NO水平,增加内皮依赖性血管舒张功能,保护血管内皮。Objective To observe the effect of Yangxin Decoction on vascular relaxation function in rats with endothelial injury induced by Hey.Methods Forty male SD rats were randomly divided into blank group,model group,Yangxin Decoction group and folic acid group with 10 rats in each group.The blank group was fed with normal diet and the other four groups were fed with diet containing 3%methionine for 12 weeks to replicate the vascular endothelial injury model of hyperhomocysteinemia.At the 13th week,all rats were fed with routine diet and given drug intervention.Rats in model group and blank group were given routine distilled water of equal volume;rats in Yangxin Decoction group were given Yangxin Decoction extract 2.34 g/kg/d by gavage;rats in folic acid group were given folic acid 1.8 mg/kg/d by gavage for 4 weeks.At the 16th weekend,rats were anesthetized and weighed.The morphological changes of endothelium of thoracic aorta were observed by HE staining.The levels of NO and ET-1 in serum were measured by ELISA,and the diastolic function of thoracic aorta was measured by isolated rings.Results Compared with the model group,Yangxin Decoction could alleviate the pathological damage of rat vascular endothelium,increase the level of serum NO,decrease the level of ET-1,and improve the endothelium-dependent vasodilation function.Conclusion Yangxin Decoction can reduce ET-1 and up-regulate NO level,in crease end othelium-dependent vasodilation function and protect endothelium.
