摘  要:【目的】分析紫斑百合与栽培百合的亲缘关系,为利用紫斑百合改良栽培百合杂交育种中亲本的选配提供指导。【方法】以6份紫斑百合野生资源和92份市场流行、广泛栽培的百合品种为材料,通过两轮温度梯度PCR筛选引物后,利用SSR引物对DNA进行PCR扩增,并进行UPGMA聚类分析。【结果】从100对SSR引物共筛选获得15对通用性好、多态性高、扩增条带清晰的引物。15对SSR引物在98份百合样品中共检测到93.0个等位位点,平均每对引物6.2个。多态性信息含量(PIC)为0.29~0.84,平均为0.66,其中PIC高于0.50的引物有13对,属于高多态性引物。UPGMA聚类分析结果显示,98份样品聚为两大分支,分支Ⅰ包括紫斑百合、O(东方百合)杂种系和OT杂种系,表明其遗传关系较近,筛选的15对SSR引物可将这3个类群进行有效区分;分支Ⅱ包括LA杂种系、L(麝香百合)杂种系和A(亚洲百合)杂种系,不同的类群间有部分交叉,筛选的15对引物不能对其进行有效区分。【结论】筛选出的15对SSR引物可将98份百合材料分为两大分支,并可应用于分支Ⅰ中3个类群的分子鉴定。紫斑百合与OT杂种系和O杂种系的遗传关系更近,在制定紫斑百合与栽培百合杂交组合时可先从OT杂种系和O杂种系的杂交改良展开研究,特别是聚在紫斑百合附近的优良OT百合品种。【Objective】The purpose of this study was to explore the genetic relationships between Lilium nepalense and cultivated lilies to provide reference for improving parent breeding in the hybrids of cultivated lilies improved by L.nepalense.【Method】A total of 6 wild L. nepalenses and 92 cultivars with market interests were analyzed in this study.SSR primer pairs were screened for polymorphism in two rounds of temperature gradient PCR. Following all samples were amplified by selected SSR primers,UPGMA cluster was performed.【Result】There were 15 SSR primer pairs with good versatility,high polymorphism and clear amplification bands being screened from 100 pairs of SSR orimers. A total of 93 allelic loci were detected in 15 primer pairs among the 98 lily samples,with an average of 6.20 alleles of every primer pair. The variation range of polymorphism information content(PIC)was 0.29-0.84,with an average value of 0.66,and PIC of 13 primer pairs was higher than 0.50,which were high polymorphism primers. UPGMA cluster analysis showed that 98 samples were divided into two main branches. BranchⅠ included L.nepalense,O(Oriental)hybrids and OT hybrids,indicating that their phylogenetic relationships was closely related,and three groups could be distinguished effectively by the selected 15 primer pairs. Branch Ⅱ included LA hybrids,L(Easter)hybrids and A(Asia)hybrids. But there were overlaps among the groups,the 15 SSR primer pairs could not classified them effectively.【Conclusion】The selected 15 SSR primer pairs can divide 98 lily samples into two main branches,and can be applied to molecular identification of three groups in branch Ⅰ. L. nepalense has a closer relationship with O hybrids and OT hybrids. When making a crossbreeding program between L. nepalense and cultivated lilies,O hybrids and OT hybrids can be given a priority to cross with L. nepalense,especially the excellent OT hybrids,which are clustered around L. nepalense.
