摘  要:WRKY转录因子是植物信号网络中不可缺少的部分,是最大的调节子家族之一。本研究从中间锦鸡儿干旱转录组及基因表达检测数据中,挑选出对干旱、冷、盐、碱及ABA都响应明显的CiWRKY12基因,对其进行克隆及生物信息学分析。结果显示:CiWRKY12基因的开放阅读框为696 bp,编码232个氨基酸,含有一个典型的WRKY保守结构域,属于GroupⅡc亚族。对转录因子CiWRKY12与其它8种植物的同源蛋白的理化性质和一、二、三级结构进行生物信息学比较分析,发现这9个蛋白无规则卷曲比例较高,大部分属于不稳定蛋白;亚细胞定位预测表明大部分的蛋白定位于细胞核内;多重序列比对显示WRKY保守结构域保守性很高,其三级结构上也呈现较高的相似性,推测这9个转录因子可能具有相似的转录调控功能。通过本研究生物信息学分析将有助于对CiWRKY12深层次功能研究提供借鉴作用。WRKY transcription factor is an indispensable part of plant signaling network and one of the largest regulators.The CiWRKY12 gene,which responds to drought,cold,salt,alkali and ABA stress,was selected from the data of drought transcriptome and gene expression detection of Caragana intermedia,and cloned and analyzed by bioinformatics.The results showed that the open reading frame of CiWRKY12 gene was 696 bp,encoding 232 amino acids.It contained a typical conserved domain of WRKY,belonging to Group IIc subgroup.Comparing the physical and chemical properties of CiWRKY12 with the homologous proteins of eight other plants,we found that the ratio of random coil of the nine proteins was comparably high,and most of them belonged to unstable proteins.Subcellular localization prediction indicated that most of the proteins were located in the nucleus.Multiple sequence alignment showed that the conserved domain of WRKY was highly conserved.The tertiary structure also shows a high similarity.It is speculated that these nine transcription factors may have similar transcriptional regulation functions.The bioinformatics analysis in this study will be helpful for the further functional study of CiWRKY12.
