摘 要:以露薇花种子无菌播种苗和穴盘播种苗为试验材料,比较两种条件下种子发芽情况,并将两种幼苗作为外植体,移入初代培养基内进行诱导培养和继代增殖。运用主成分分析法和相关性分析法,并通过建立拟合生长曲线,量化评价不同播种方式幼苗离体培养下生长情况。结果表明,穴盘播种发芽率较无菌播种发芽率高,幼苗根系发达,但苗离体培养污染率较高;离体无菌培养更有利于露薇花植株生长发育的进行,可将穴盘发芽幼苗灭菌后移植进培养基培养;继代培养穴盘播种和无菌播种苗之间无差异;继代培养3次,单瓶接种两株植株,植株长势最好,萌芽数,萌芽均高,增殖倍数高。通过本次实验,为露薇花高效生产繁殖提供理论依据,从而为西北地区新优特异花卉的引进,提供可行的途径方法。Seed germination was compared between sterile seedling and hole seedling of Lewisia cotyledon under two conditions.The two seedlings were used as explants and transferred into the primary culture medium for induction culture and subculture.Principal component analysis and correlation analysis were used to quantify the growth of seedlings in different seeding cultures by establishing a fitted growth curve.The results showed that the germination rate of the seeding tray was higher than that of the aseptic seeding,and the seedling roots were developed,but the seedling culture rate was higher.In vitro sterile culture is more conducive to the growth and development of the Lewisia cotyledon plant,and the seedling sprouting seedlings can be sterilized and transplanted into the medium culture.There was no difference between subcultured tray seeding and aseptic seeding.Subculture was carried out 3 times,and two plants were inoculated in a single bottlei.The plant growth wais the best,the number of germination was high,the budding average height was high,and the multiplication ratio was high.Through this experiment,we can provide theoretical basis for the efficient production and reproduction of Lewisia cotyledon,and provide feasible ways and means for the introduction of new and special flowers in Northwest China.
