摘  要:目的研究银屑病角质形成细胞核因子κB抑制剂α(nuclear factor-κB inhibitorα,NFKBIA)的表达。方法收集36例银屑病患者皮损组织及30例正常切色素痣者的痣旁正常组织,通过实时荧光定量聚合酶链式反应法(quantitative real time polymerasechainreaction,q RT-PCR)检测组织中白介素(interleukin,IL)-17和NFKBIA的表达。同时,培养正常人原代角质形成细胞,以100 ng/ml IL-17干预24 h,q RT-PCR法检测细胞CC趋化因子配体20(C-C motif chemokine ligand 20,CCL20)、IL-8、β-防御素1(β-defensin 1,HBD1)、S100钙结合蛋白A7(S100calciumbindingprotein A7,S100A7)的表达。分别以100、200及300 ng/ml浓度的IL-17干预原代角质形成细胞24 h,q RT-PCR法检测细胞NFKBIA的表达。100 ng/ml IL-17干预原代角质形成细胞0、10、20、30及60 min,免疫印迹法(immunoblot,western-blot)检测细胞NFKBIA蛋白的表达。结果在银屑病皮损组织中,IL-17表达高于正常组织(P <0.01),而NFKBIA表达低于正常组织(P <0.05)。IL-17干预原代角质形成细胞24 h后CCL20、IL-8、HBD1、S100A7等炎症因子表达明显升高,差异均有统计学意义(P <0.01)。不同浓度IL-17干预原代角质形成细胞24 h后,NFKBIA信使RNA表达无显著性差异(P> 0.05),NFKBIA的表达与IL-17浓度无明显梯度关系。100 ng/ml IL-17干预人原代角质形成细胞30及60 min时NFKBIA蛋白表达下降。结论 NFKBIA在银屑病角质形成细胞中表达降低,可能与IL-17促进银屑病角质形成细胞炎症有关。Objective To investigate the expression of nuclear factor-κB inhibitor α(NFKBIA) in the keratinocytes of psoriasis.Methods The expressions of interleukin 17(IL-17) and NFKBIA in 36 cases of psoriasis samples and 30 cases of normal samples closes to the nevi were detected by quantitative real time polymerase chain reaction(qRT-PCR).The expressions of C-C motif chemokine ligand 20 (CCL20),IL-8,β-defensin 1(HBD1) and S100 calcium binding protein A7(S100A7) in the primary keratinocytes culture by the treatment with 100 ng/ml IL-17 for 24 h were detected by qRT-PCR.The expression of NFKBIA in the primary keratinocytes culture by the treatment with different concentrations of IL-17 for 24 h were detected by qRT-PCR.The expression of NFKBIA protein in the primary keratinocytes by the treatment with 100 ng/ml IL-17 for 0,10,20,30 and 60 min were detected by immunoblot(western blot).Results The expression of IL-17 was increased(P<0.01),while the expression of NFKBIA was decreased(P<0.05) in psoriasis samples.The expressions of CCL20、IL-8、HBD1 and S100A7 in the primary keratinocytes were significantly higher than the blank controls after the treatment with 100 ng/ml IL-17 for 24 h(P<0.01).There was no significant difference in NFKBIA mRNA expression after treatment with different concentrations of IL-17 for 24 h(P>0.05),which was no related with the concentration of IL-17.But the expression of NFKBIA protein was obviously decreased in the keratinocytes treated by 100 ng/ml IL-17 at the time of 30 and 60 min.Conclusion The expression of NFKBIA was decreased in psoriasis keratinocytes,which may be related with the inflammation production of keratinocytes treated by IL-17.
