摘  要:目的评估鼻黏膜组织微小RNA(miR)-103和miR-107表达量与变应性鼻炎(AR)患病风险、疾病严重程度和炎症因子表达水平的关联。方法纳入80例AR患者和80例年龄、性别匹配的对照组,获取其鼻黏膜组织并采用定量聚合酶链式反应检测miR-103,miR-107,肿瘤坏死因子α(TNF-α),白细胞介素(IL)-1β,IL-4,IL-6,IL-13,IL-8,IL-10,IL-17 mRNA的表达水平。评估AR患者单个鼻症状评分(INSS)和总鼻症状评分(TNSS)。结果AR患者鼻黏膜组织的miR-103和miR-107表达量均低于对照组(P均<0.001)。ROC曲线分析显示,鼻黏膜组织的miR-103表达量可以在一定程度上预测AR患病风险,鼻黏膜组织的miR-107表达量对AR患病风险具有较好的预测作用,其AUC分别为0.752(95%CI:0.678~0.826)和0.886(95%CI:0.834~0.939)。鼻黏膜组织的miR-103和miR-107表达量在AR患者(P<0.001)和对照组(P<0.001)均呈正相关。AR患者鼻黏膜组织的miR-103和miR-107表达量与鼻痒评分、鼻塞评分和TNSS均呈负相关(P均<0.05),与促炎因子表达水平呈负相关,与抑炎因子水平呈正相关(P均<0.05);对照组鼻黏膜组织的miR-103和miR-107表达水平也与促炎因子表达量呈负相关(P均<0.05)。结论鼻黏膜的miR-103和miR-107在AR患者中低表达,且与AR的患病风险、疾病严重程度和炎症水平呈负相关。Objective To evaluate the correlations of nasal mucosa microRNA(miR)-103 and miR-107 expressions with risk,severity,and inflammatory cytokines levels of allergic rhinitis(AR).Methods 80 AR patients and 80 age and sex matched controls were enrolled in this study.The nasal mucosa tissue samples were obtained from all the AR patients and controls for the detection of miR-103,miR-107,tumor necrosis factor(TNF)-α,and interleukin(IL)-1β,IL-4,IL-6,IL-13,IL-8,IL-10,IL-17 mRNA expressions by quantitative Polymerase Chain Reaction(qPCR).In addition,individual nasal symptom score(INSS)and total nasal symptom score(TNSS)of AR patients were recorded.Results The nasal mucosa miR-103 expression and miR-107 expression were downregulated in AR patients compared with controls(all P<0.001).The ROC curve analysis revealed that nasal mucosa expression of miR-103 could predict the AR risk to some extent,and miR-107 had good value in predicting AR risk,with AUC values of 0.752(95%CI:0.678-0.826)and 0.886(95%CI:0.834-0.939),respectively.And nasal mucosa miR-103 expression was positively correlated with miR-107 expression in both AR patients and controls(all P<0.05).In AR patients,nasal mucosa miR-103 and miR-107 expressions were negatively correlated with itching score,conjunction score and TNSS score(all P<0.05).In addition,nasal mucosa miR-103 and miR-107 expressions were negatively correlated with pro-inflammatory cytokines levels and were positively associated with anti-inflammatory cytokine level(all P<0.05)in AR patients.In controls,nasal mucosa miR-103 and miR-107 expressions were also positively associated with pro-inflammatory cytokines expressions.Conclusion Nasal mucosa miR-103 and miR-107 expressions are downregulated in AR patients,and are negatively correlated with AR risk,severity and inflammation level.
