益气除湿活血汤对肥胖大鼠糖、脂代谢的影响

摘　　要：目的:研究中药益气除湿活血汤对肥胖大鼠糖、脂代谢的影响及其分子机制。方法:40只SD大鼠随机分为对照组、肥胖组、肥胖+益气汤组和肥胖+二甲双胍组,每组各10只。以高脂、高糖饲料喂养大鼠12周制备肥胖大鼠模型,对照组大鼠给予基础饲料喂养。肥胖+益气汤组、肥胖+二甲双胍组和肥胖组动物分别给予8周益气除湿活血汤、二甲双胍、生理盐水灌胃处理。测定动物体质量并计算Lee′s指数,取血测定空腹血糖(FBG)及血胆固醇和三酰甘油浓度、胰岛素抵抗指数(IRI)、肿瘤坏死因子-α(TNF-α)和脂联素水平。结果:与对照组大鼠相比较,肥胖组大鼠体质量、Lee′s指数、FBG、总胆固醇、三酰甘油和IRI均显著升高(P<0.05,P<0.01),血清脂联素明显降低(P<0.01),TNF-α明显升高(P<0.01)。与肥胖组大鼠相比较,肥胖+二甲双胍组大鼠体质量、Lee′s指数、FBG和IRI明显降低(P<0.05,P<0.01),脂联素水平明显升高(P<0.05);肥胖+益气汤组大鼠体质量、Lee′s指数、FBG、IRI、总胆固醇和三酰甘油明显降低(P<0.05)外,血清TNF-α明显降低,脂联素明显升高(P<0.05,P<0.01)。结论:益气除湿活血汤可改善肥胖大鼠糖、脂代谢紊乱和胰岛素抵抗,此作用可能通过升高血脂联素、降低TNF-α所实现。Objective:To study the effect of Yiqi Chushi Huoxue Decoction on glucose and lipid metabolism in obese rats and its molecular mechanism.Methods:40 SD rats were randomly divided into control group,obesity group,obesity&Yiqi Decoction group and obesity&metformin group with 10 rats in each group.The obese rats were fed with high fat and high sugar diet for 12 weeks,and the control rats were fed with basic diet.The obese&Yiqi Decoction group,obese&metformin group and obese group were given Yiqi Chushi Huoxue Decoction,metformin and normal saline respectively for 8 weeks.The body mass of the animals was measured and theLee′s index was calculated.The fasting blood glucose(FBG),serum cholesterol and triglyceride concentration,insulin resistance index(IRI),tumor necrosis factor-α(TNF-α)and adiponectin levels were measured.Results:Compared with the control group,the body weight,Lee′s index,FBG,total cholesterol,triacylglycerol and IRI of the obese group increased significantly(P<0.05,P<0.01),serum adiponectin decreased significantly(P<0.01),TNF-αincreased significantly(P<0.01);compared with the obese group,the body weight,Lee′s index,FBG and IRI of the obese&metformin group decreased significantly(P<0.05,P<0.01);The body weight,Lee′s index,fasting blood glucose,IRI,total cholesterol and triacylglycerol were significantly decreased in obesity&Yiqi Decoction group(P<0.05),while TNF-αwas significantly decreased and adiponectin was significantly increased(P<0.05,P<0.01).Conclusion:Yiqi Chushi Huoxue Decoction can improve the disorder of glucose and lipid metabolism and insulin resistance in obese rats,which may be achieved by increasing adiponectin and reducing TNF-α.
