摘  要:目的探讨采用胫骨局部处用微量加样器皮下接种注射Ⅰ型单纯疱疹病毒(HSV-Ⅰ)制作稳定、可靠的带状疱疹后神经痛(PHN)小鼠模型的方法。方法将8周龄雄性C57小鼠30只随机分为3组,分别为空白组、实验组和对照组,每组10只。实验组分别于每只小鼠左下肢胫骨局部处用微量加样器皮下接种HSV-Ⅰ10μL(浓度为1×106 PFU/mL),对照组以同等剂量的灭活HSV-Ⅰ进行接种,空白组在相应部位注射同等剂量的生理盐水。在造模前(T 0)和造模后1 d(T 1)、3 d(T 2)、5 d(T 3)、7 d(T 4)分别测定小鼠后爪机械缩足反应阈值(MWT)和热痛阈值(TWL)。结果实验组在T 2~4时的MWT均显著性降低,与空白组及对照组比较,差异均有统计学意义(P<0.05);在T 4时MWT达到最低[(1.42±0.22)g]。实验组在T 1~4时的TWL均显著性升高,与空白组及对照组比较,差异均有统计学意义(P<0.05);在T 4时TWL达到最高[(17.23±0.70)s]。实验组造模成功率为100%。结论于胫骨局部处用微量加样器皮下接种注射成功诱导小鼠PHN模型是可行的,并贴近疾病发展规律,具有造模成功率高、损伤小、操作简单便捷、可重复性高等优点。Objective To study the method of making a stable and reliable model of postherpes neuralgia mice by subcutaneous injection with herpes simplex virusⅠ(HSV-Ⅰ)with microspline injector in tibia.Methods Thirty male C57 mice aged 8 weeks were randomly divided into three groups:blank group,experimental group and control group,each group of 10.The experimental group inoculated each mouse with a microspline subcutaneous HSV-Ⅰ10μL(concentration 1×106 PFU/mL)at the local area of the left lower limb of the tibia.The control group was inoculated with the same dose of inactivated HSV-Ⅰ.The blank group injected the same dose of saline at the corresponding site.The me-chanical withdraw threshold(MWT)and heat pain threshold(TWL)were measured before(T 0)and 1 d(T 1),3 d(T 2),5 d(T 3)and 7 d(T 4),respectively.Results The MWT at the T 2-4 period were significantly reduced in the experimental group,and compared with the blank group and the control group,the differences were statistically significant(P<0.05),and reached the lowest threshold at the T 4[(1.42±0.22)g].The TWL at the T 1-4 period were significantly reduced in the experimental group,and compared with the blank group and the control group,the differences were statistically significant(P<0.05),and reached the highest threshold at T 4[(17.23±0.70)s].The modeling success rate of the experimental group was 100%.Conclusion It is feasible to induce the mouse PHN model successfully with microspline subcutaneous inoculation in tibia,and it is close to the disease development law.It has the advantages of high success rate,small damage,simple and convenient operation,and high repeatability.
