摘  要:目的:观察健脾养血汤剂对特异性皮炎小鼠模型皮肤炎症症状和Toll样受体表达的影响。方法:通过外用2,4-二硝基氟苯(DNFB)丙酮制备特异性皮炎小鼠模型,小鼠随机分为正常对照组(n=10)、模型组(n=10)和健脾养血汤剂治疗组(n=10)。造模后10d各组皮炎症状打分,使用实时荧光定量聚合酶链反应检测各组皮肤组织中TLR2、TLR3、TLR4、TLR9m RNA表达。结果:与正常组比较,模型组和健脾养血汤治疗组皮炎症状显著,TLR2、TLR3、TLR4、TLR9m RNA均明显上升,表达显著增高(P<0.05)。与模型组比较,健脾养血汤治疗组皮炎症状显著减轻,TLR2、TLR3、TLR4、TLR9m RNA表达均降低(P<0.05)。结论:健脾养血汤剂可通过抑制TLR2、TLR3、TLR4、TLR9表达,改善特异性皮炎临床症状。Objective:To observe the effects of Jianpi Yangxue Decoction on skin inflammatory symptoms and Toll-like receptors expression in the mouse model of atopic dermatitis.Methods:The model of atopic dermatitis was established in mice by applying of 2,4-dinitrofluorobenzene(DNFB)to the skin,the mice were randomly divided into normal control group(n=10),model group(n=10)and decoction observation group(n=10).The inflammatory symptoms on skin of each group were scored 10 days after model establishment,and the expression of TLR2,TLR3,TLR4 and TLR9 mRNA in the skin of each group were detected by real-time quantitative PCR.Results:Compared with the normal group,the dermatitis symptoms in the model group and decoction observation group were significant,and the expression of TLR2,TLR3,TLR4,and TLR9 mRNA were significantly increased(P<0.05).Compared with the model group,Jianpi Yangxue Decoction significantly reduced the symptoms of dermatitis,and decreased the expression of TLR2,TLR3,TLR4 and TLR9 mRNA(P<0.05).Conclusion:Jianpi Yangxue Decoction can alleviate the clinical symptoms of atopic dermatitis by inhibiting the expression of TLR2,TLR3,TLR4 and TLR9.
