摘  要:目的:建立多疣壁虎中枢神经系统结构的三维立体模型,获取中枢神经系统相关结构的定量数据。方法:多疣壁虎头颈部连续冠状切片行Nissl染色和Luxol fast blue(LFB)染色,通过显微镜分区拍摄获得切片的二维图像。利用Photoshop7.0对图像拼接、旋转、配准后,采用3D-Doctor软件进行三维重建。运用体视学方法对壁虎头部进行定量研究。结果:三维重建的头颅、脑和视神经的立体图像逼真自然,能清楚地区分端脑、间脑、中脑、延髓以及小脑,同时能弄清壁虎脑在颅内位置及相互关系。壁虎颅腔的容积为(25.83±1.46)mm^3,全脑体积为(19.99±2.15)mm^3、端脑体积为(7.53±0.33)mm^3、单侧大脑皮层体积为(0.19±0.02)mm^3、小脑体积为(0.22±0.02)mm^3、海马体积为(0.16±0.02)mm^3,脊髓颈膨大、灰质和白质面积分别为(0.70±0.02)mm^2、(0.28±0.01)mm^2、(0.36±0.01)mm^2,颈膨大灰质与白质面积比是0.78,腰骶膨大、灰质和白质的面积分别为(0.75±0.01)mm^2、(0.36±0.01)mm^2、(0.38±0.01)mm^2,腰骶膨大灰质与白质面积比是0.95。结论:获得多疣壁虎颅脑解剖结构的三维立体模型及相关脑结构的定量分析数据,为壁虎脑内实验和立体定向研究提供了三维解剖学依据。Objective:To build the three-dimensional model of central nervous system(CNS)structure of Gekko japonicus and obtain the quantitative data of these structures of Gekko.Methods:Based on the Nissl's and Luxol fast blue(LFB)staining,the image of serial coronal sections of the head and truncus of Gekko japonicus were obtained by zone shooting using microscope.The 3D model was reconstructed by 3D-Doctor software after splicing,rotating and rectificating the images with Photoshop 7.0.Then the head of Gekko japonicus was investigated quantitatively by the stereological method.Results:Three-dimensional stereoscopic images of the skull,head and optic nerve were realistic and natural,from which the telencephalon,diencephalon,midbrain,medulla and cerebellum could be clearly distinguished.The cranial cavity of the Gekko japonicus was(25.83±1.46)mm^3,the whole volume of encephalon was(19.99±2.15)mm^3,the volume of the telencephalon was(7.53±0.33)mm^3,the volume of unilateral cerebral cortex was(0.19±0.02)mm^3,the volume of cerebellum was(0.22±0.02)mm^3,the volume of hippocampus was(0.16±0.02)mm^3,the area of the cervical segment of spinal cord was(1.68±0.35)mm^2,and the area of the spinal gray matter of cervical segment was(0.98±0.30)mm^2.The average area of the cervical enlargement,gray matter and white matter was(0.70±0.02)mm^2,(0.28±0.01)mm^2,(0.36±0.01)mm^2,respectively,and the area ratio of gray matter and white matter was 0.78;the average area of the lumbosacral enlargement,gray matter and white matter was(0.75±0.01)mm^2,(0.36±0.01)mm^2,(0.36±0.01)mm^2,respectively,and the area ratio of gray matter and white matter was 0.95.Conclusion:The 3D model and the quantitative analysis of anatomical structure of the brain of Gekko japonicus can provide 3D anatomical data for the intracerebral experiment and stereotactic research.
