摘 要:目的:观察消渴平合剂对db/db小鼠肾脏P38MAPK-FN信号通路表达的影响,探讨其防治糖尿病肾病的机制。方法:24只db/db小鼠,按随机数字表法分为模型组、消渴平组,厄贝沙坦组,每组各8只,另取8只db/m小鼠为正常对照组,连续给药8周后分别检测各组小鼠体质量、血肌酐、尿素氮。留取肾组织,采用Western blot检测肾脏P38MAPK、FN蛋白的表达、荧光定量PCR检测肾脏P38MAPKmRNA、FNmRNA的表达。结果:消渴平合剂可以明显降低db/db小鼠体质量及血肌酐、尿素氮水平。与正常对照组相比,模型组小鼠肾脏P38MAPK、FN的表达显著升高(P<0.01);与模型组相比,消渴平组可明显抑制P38MAPK、FN的表达(P<0.05)。结论:消渴平合剂可明显减轻db/db小鼠肾脏损伤,其机制可能与抑制肾脏P38MAPK-FN信号通路激活有关。Objective:To observe the effects of Xiaokeping Mixture on the expressions of P38MAPK-FN signaling pathway in the kidney of db/db mice,and to explore the mechanism of its prevention of diabetic kidney disease.Methods:24 db/db mice were divided into model group,Xiaokeping group and Irbexatan group by random number table method with 8 mice in each group,another 8 db/m mice were taken as the normal control group.Body weight,SCr and urea nitrogen were measured after 8 weeks of continuous administration.Kidney tissues were collected and the expressions of P38MAPK and FN proteins in the kidney were detected by Western blot,and the expressions of P38MAPK and FNmRNA in the kidney were detected by fluorescence quantitative PCR.Results:Xiaokeping Mixture could significantly reduce the body mass,SCr and urea nitrogen levels in db/db mice.Compared with the normal control group,the expressions of P38MAPK and FN in the kidney of the model group were significantly increased(P<0.01).Compared with the model group,the expressions of P38MAPK and FN were significantly inhibited in the Xiaokeping group(P<0.05).Conclusion:Xiaokeping Mixture can significantly reduce renal injury in db/db mice,which may be related to the inhibition of renal P38MAPK-FN signal pathway activation.
