摘 要:目的:研究四物汤对兔受损神经节组织形态学变化和细胞学凋亡的影响。方法:将24只新西兰大白兔随机分为假手术组、模型组、四物汤组,每组8只。以铬制肠线环扎背根神经造模,四物汤组予四物汤4.68 g/(kg·d)灌胃给药;96 h后,观察各组兔背根神经组织水肿和粘连状况,并行石蜡包埋切片、HE染色观察病理变化和TUNEL法染色观察细胞凋亡,计算凋亡指数(AI)。结果:假手术组兔神经节水肿及与周围组织粘连不明显;模型组可见神经节水肿,与周围组织粘连;四物汤组神经节水肿程度较低,与周围组织轻度粘连。HE染色可见:假手术组神经节组织细胞病理学改变不明显;模型组神经节细胞间隙宽且皱缩,细胞排列欠规则,尼氏小体减少,细胞核缩小;四物汤组神经节细胞间隙增宽不明显,细胞排列较规则。TUNEL法染色可见:模型组神经节细胞凋亡率明显高于假手术组,四物汤组神经节细胞凋亡率低于对照组,但高于假手术组(P<0.05)。结论:铬制肠线环扎背根神经可诱发兔背根神经损伤;四物汤可以改善受损神经节组织形态病理变化,抑制受损神经节细胞衰亡。Objective:To study the effects of Siwu Decoction on morphological changes and apoptosis of injured ganglion in rabbits.Methods:24 New Zealand white rabbits were randomly divided into sham operation group,model group and Siwu Decoction group,8 in each group.The model was made by encircling the dorsal root nerve with chrome gut,Siwu Decoction group was given 4.68 g/(kg·d)Siwu Decoction by gavage;96 hours later,the edema and adhesion of the dorsal root nerve tissue were observed,Paraffin embedded sections,HE staining and TUNEL staining were used to observe apoptosis of injured ganglion cells;calculated the apoptosis index(AI).Results:In the sham operation group,there was no obvious swelling and adhesion with the surrounding tissue;in the model group,there was ganglioedema and adhesion with the surrounding tissue;in the Siwu Decoction group,the degree of ganglioedema was low and slightly adhesion with the surrounding tissue.HE staining showed that the pathological changes of ganglion cells in sham operation group were not obvious;the gap of ganglion cells in model group was wide and shrunk,the arrangement of cells was not regular,the number of Nissl bodies was reduced,and the nucleus was reduced;the gap of ganglion cells in Siwu Decoction group was not obvious,and the arrangement of cells was more regular.TUNEL staining showed that the apoptosis rate of ganglion cells in the model group was significantly higher than that in the sham operation group,and that in the Siwu Decoction group was lower than that in the control group,but higher than that in the sham operation group(P<0.05).Conclusion:Chromium gut can induce dorsal root nerve injury,Siwu Decoction can improve the morphological and pathological changes of the injured ganglion and inhibit the decline of the injured ganglion cells.
