摘 要:目的:观察白藜芦醇(Res)对抑郁小鼠脑组织mir-155相关炎症因子表达的影响。方法:采用慢性不可预见轻度应激方法建立抑郁小鼠模型。将抑郁小鼠随机分为模型组、Res(20、40 mg/kg)组和氟西汀组,每组8只。观察抑郁小鼠糖水消耗指数、强迫游泳不动时间和新奇摄食时间,实时定量PCR检测脑组织mir-155相关炎症因子TNF-α(肿瘤坏死因子)、IL-6(白细胞介素-6)和吲哚胺2,3-双加氧酶(IDO)mRNA表达水平;免疫印迹法分析TNF-α、细胞因子信号抑制物1(SOCS1)和IDO蛋白的表达水平。结果:Res能够升高抑郁小鼠糖水消耗指数、缩短强迫游泳不动时间和新奇摄食时间,下调脑组织mir-155、TNF、IL-6和IDO表达,上调SOCS1表达。结论:Res能够改善抑郁小鼠行为学异常,其作用机制与抑制mir-155参与的中枢炎症有关。Objective:To explore the effects of Resveratrol on expressions of mir-155 related inflammatory factors in depress mice.Methods:The chronic unpredictable stress mice model with depression was employed in this study.The depression mice were randomly divided into four groups:depression mice,Res(20,40 mg/kg)and Fluoxetine.Sugar and water consumption index,time of motionlessness in forced swimming test and novelty feeding test were observed.The mRNA expressions of mir-155 related inflammation factors and IDO wrer determined with Real-time PCR.The protein expressions of SOCS1,TNF-αand IDO were determined with Western blot.Results:Res could increase the sugar and water consumption index,shorten the time of motionlessness in forced swimming test and novelty feeding test of depression mice.Additionally,Res decreased the mRNA expressions of mir-155,TNF-αIL-6 and IDO,and elevated the expression of SOCS1.Conclusion:Res could improve behavioral abnormalities of depression mice,the mechanism is relevant with the inhibition of mir-155 related inflammation of central nervous system.
