摘  要:目的:分析远华蟾毒精对于MMP-2、MMP-9的合成相关信号传导通路抑制的作用与机制及对乳腺癌细胞增殖作用的影响。方法:将培养好的裸鼠乳腺癌抑制瘤组织切片均分为3组。A组仅使用常规培养;B组在培养过程中加入0.1μmol/L远华蟾毒精;C组在培养过程中加入0.5μmol/L远华蟾毒精。对三组组织切片进行Western Blot检测。比较三组MMP-9、MMP-2表达情况与细胞迁移距离情况。结果:培养结束后,C组MMP-9、MMP-2表达量及不同时间点细胞迁移距离均显著低于B组与A组,差异有统计学意义(P<0.05)。结论:远华蟾毒精可有效通过抑制MMP-2及MMP-9合成,阻滞乳腺癌细胞迁移,从而达到抑制乳腺癌细胞增殖,阻滞细胞转移效果,可根据此设计相应治疗方案,为乳腺癌患者提供新的治疗手段。Objective:To analyze the inhibitory effect and mechanism of telocinobufagin on the regulating signal transduction pathways related to the synthesis of MMP-2,MMP-9 and the effect on the proliferation of breast cancer cells.Methods:The cultured nude mice breast cancer suppressor tissue sections were divided into 3 groups.Group A only used routine culture,group B added 0.1μmol/L telocinobufagin in the culture process and group C added 0.5μmol/L telocinobufagin in the culture process.Western Blot detection was performed on three groups of tissue sections.The expression of MMP-9 and MMP-2 and the distance of cell migration were compared.Results:After culture,the expression of MMP-9,MMP-2 and the migration distance of cells in group C were significantly lower than those in group B and group A at different time points,the differences were statistically significant(P<0.05).Conclusion:Telocinobufagin can effectively inhibit the migration of breast cancer cells by inhibiting the synthesis of MMP-2 and MMP-9,thereby inhibiting the proliferation of breast cancer cells and blocking cell metastasis,according to this,the corresponding treatment plan can be designed to provide new treatment methods for breast cancer patients.
