摘 要:单核苷酸多态性(SNP)是基因组中普遍存在的遗传变异,是一种重要的遗传标记。为了解拟环纹豹蛛转录组中响应温度胁迫的SNP候选位点及SNP所在基因SNP-Unigenes的功能注释,主要采用SOAPsnp软件对拟环纹豹蛛在不同温度胁迫条件下的转录组测序结果进行SNP检测,并把SNP所在基因SNP-Unigenes比对到GO、COG、KEGG数据库,获得SNP-Unigenes的注释及参与的功能情况。结果表明:拟环纹豹蛛转录组SNP位点分布在7421条SNP-Unigenes上,共存在402910个SNP位点,SNP发生频率为1/301 bp;功能注释分类揭示拟环纹豹蛛在低温胁迫和高温胁迫条件下,SNP-Unigenes主要参与蛋白质转运、核糖体代谢、氧化磷酸化和氨基酸代谢功能,从而补偿热应力下蛋白质的损伤,加快拟环纹豹蛛对温度变化的适应。综上,筛选到拟环纹豹蛛响应温度胁迫相关的SNP位点或SNP-Unigenes,为将来深入研究拟环纹豹蛛响应温度胁迫的分子机制提供基础数据。SNP is a common genetic variation in genome and an important genetic marker.In order to mine SNP related to temperature tolerance and understand the functional of the SNP-Unigenes from Pardosa pseudoannulata,SOAPsnp software was used to detect the SNP from transcriptome sequencing results of P.pseudoannulata under different temperature stress conditions,and the annotation of SNP-Unigenes in the databases of GO,COG and KEGG was compared.The results showed that the transcriptome SNP sites were distributed in 7421 SNP-Unigenes,with a total of 402910 SNP sites and the frequency of SNP occurrence was 1/301 bp.Functional annotation classification revealed that SNP-Unigenes was mainly involved in protein transport,ribosome metabolism,oxidative phosphorylation and amino acid metabolism,when spiders were under low temperature and high temperature stress,so as to compensate for protein damage under thermal stress and speed up the adaptation to temperature change of P.pseudoannulata.SNP sites or SNP-Unigenes response to temperature stress were selected,which laid a theoretical foundation for further studies on the molecular mechanism of P.pseudoannulata in response to temperature stress.
