摘  要:研究脂肪组织形成是揭示肥胖和代谢疾病发生机制的重要途径,细胞水平研究显示GATA2(GATA binding protein 2)、GATA3(GATA binding protein 3)、KLF2(Kr俟ppel-like Factor 2)、KLF3(Kr俟ppel-like factor 3)、KLF7(Kruppel-like factor 7)、MTG8(Runt-related transcription factor 1)和DLK1(Protein delta homolog 1)抑制3T3-L1细胞向脂肪细胞的分化。但是,目前这7个基因在脂肪组织形成过程中表达间的相互关系还不完全清楚。本研究利用半定量RT-PCR研究了这7个基因在鸡腹部脂肪组织生长发育过程中的表达情况。显示上述7个基因在鸡腹部脂肪组织生长发育的所有检测时间点均有表达,相关性分析显示多个基因的表达数据间存在显著相关(p<0.05),特别是GATA2的表达数据和其他6个基因表达数据均显著正相关(p<0.05),提示了脂肪组织形成过程中这7个基因可能存在功能上的联系。主成分分析从这7个基因的表达数据中提取出了1个主成分(Eigenvalue=3.4099,Proportion=0.4871)。因子分析显示在该主成分中GATA3、KLF3和KLF7的贡献率大于0.5,GATA2和KLF2的贡献率接近0.5,DLK1和MTG8的贡献率较低。聚类分析显示,这7个基因的表达模式大致分为两类,一类为GATA2、GATA3、KLF2和KLF7,另一类为KLF3、DLK1和MTG8。本研究为进一步研究脂肪组织形成的分子调控机制提供参考。A detailed understanding of the processes governing adipose tissue formation will be instrumental in combating the obesity and metabolic disease. GATA2, GATA3, KLF2, KLF3, KLF7, MTG8 and DLK1 inhibited the adipogenesis of 3 T3-L1 cells. However, the correlation among these 7 genes in the process of adipose tissue formation were still not clearly elucidated. In the present study, semi-quantitative RT-PCR method was employed to analyze the expression of these 7 genes in the abdominal adipose tissue of chicken. It showed that all the expression of these 7 genes could be detected in the chicken abdominal adipose tissue at the time points analyzed(2 to 12 weeks of age, each time point, n=6). Correlation analysis showed that significant associations were existed among several genes(p<0.05), and the expression data of GATA2 was significantly associated with those of other 6 genes(p<0.05). Principal component analysis extracted one component from these 7 gene expression data(Eigenvalue=3.409 9, Proportion=0.487 1). Factor analysis showed thatGATA3, KLF3, KLF7, GATA2 and KLF2 contributed great power to this component. Cluster analysis showed that the expression data of these seven genes were divided into two groups based on the cluster number of 1.5, and one group included GATA2, GATA3, KLF2 and KLF7, the other group contained KLF3, DLK1 and MTG8. The present study might be shed a light on the further understanding of molecular mechanism of adipose tissue formation.
