摘 要:为查明崇明白山羊群体的遗传多样性水平和亲源关系,应用ISSR分子标记技术,使用12条多态性较好的ISSR引物对206只崇明白山羊进行PCR扩增。结果表明:总计扩增出181条谱带,平均每条引物扩增产生15.08条,其中含有多态性条带163条;经统计分析,崇明白山羊呈现出丰富的多样性,检测到的观测等位基因数(Na)为1.9006±0.3001,有效等位基因数(Ne)为1.4778±0.3656,Nei’s基因多样度(h)为0.2785±0.1849,多态位点百分率(PPB)为90.06%,Shannon’s信息指数(I)为0.4186±0.2510,群体总基因多样性(Ht)为0.2797±0.0340,群体内基因多样性(Hs)为0.2267±0.0258,遗传分化值(Gst)为0.1894,崇明白山羊的遗传变异81.06%来源于种群内部。崇明白山羊的基因流(Nm)为2.1397,说明崇明白山羊群体间存在一定的基因流。UPGMA聚类分析显示:样本相似系数为0.5193~0.9890,206个个体可分为11个群体,个体间虽然呈现差异,但具有较好的同质性。The aim of this study was to investigate the population genetic diversity and genetic relationship of Chongming white goatby molecular ISSR markers,12 screenedpolymorphic ISSR primers were used for the PCR amplification of 206 Chongming white goats.A total of 181 bands,including 163 polymorphic bands,with an average of 15.08 bands per primer were got in this study.This results showed that Chongming white goat had a rich diversity,the observed number of alleles(Na)was 1.9006±0.3001,effective number of alleles(Ne)was 1.4778±0.3656,Nei’s gene diversity(H)was 0.2785±0.1849,percentage of polymorphic loci(PPB)was 90.06%,Shannon’s information index(I)was 0.4186±0.2510,total population genetic diversity(Ht)was 0.2797±0.0340,gene diversity within populations(Hs)was 0.2267±0.0258,value of genetic differentiation(Gst)was 0.1894,and 81.06%genetic variation of Chongming white goat was from the interior of population,thegene flow(Nm)of Chongming white goat was 2.1397,which indicated that there was a certain gene flow between the different groups of Chongming white goat.UPGMA cluster analysis showed that the similarity coefficient of was 0.5193—0.9890,206 individuals could be divided into 11 groups,and although there were some differences among individuals,they had a good homogeneity.
