小剂量预适应对京尼平苷致大鼠急性肝肾损伤的影响

摘　　要：目的为含栀子及京尼平苷类药物的非临床评价和临床应用风险评估提供科学依据。方法灌胃给药4d后,观察大鼠外观、行为及体重变化;称量肝肾重并计算脏器指数;测定大鼠血清中ALT、AST的活性以及UREA、CRE的含量。结果与空白组比较,各给药组肝重及肝重指数显著增加,ALT、AST活性升高;而肾重及肾重指数与UREA、CRE的含量呈下降趋势。在小剂量京尼平苷干预后,可降低肝重指数及ALT、AST的活性,而对肾重指数及UREA、CRE的含量无降低作用。结论小剂量预适应对京尼平苷和京尼平致大鼠肝损伤具有改善作用。Objective To provide a scientific basis for the non-clinical evaluation and the assessment of clinical application risk of drugs containing Fructus Gardeniae and geniposide.Methods Observe the change of appearance,behavior and weight of rats through oral gavage daily for 4 d.Weigh the liver and kidney and calculate the liver and kidney index.Detect the activity of ALT,AST and the content of UREA,CRE to indicate the toxicity of liver and kidney injury.Results Compared with the normal control group,the weight and index of liver are markedly increased by the overdosage of geniposide or genipin;The activities of ALT and AST are also increased.But the weight and index of kidney and the content of UREA,CRE reduce.Under the interference of small dose of geniposide,the weight and index of liver slightly reduce,the activities of ALT and AST also slightly reduce.But it had no effect on the kidney index and the content of UREA,CRE.Conclusion Geniposide(or genipin)-induced hepatotoxicity is reduced under the interference of small dose of geniposide.
