摘 要:以张家口地道性中药材金莲花为试材,研究4℃低温、不同甘露醇浓度、不同蔗糖浓度在不同时间下对金莲花花药诱导愈伤组织的影响。结果表明:4℃低温处理以2 d的效果最好,诱导率为85%,显著高于未进行低温预处理的花药;甘露醇处理以浓度为0.3 mol/L时处理3 d最优;诱导率达到80.25%,显著高于其它浓度甘露醇处理及对照;蔗糖处理以0.1 mol/L浓度浸泡金莲花花蕾60 min为最佳,诱导率为92.16%,显著高于其它浓度处理及对照。说明不同预处理方式都对金莲花愈伤组织的形成有一定促进作用,但以0.1 mol/L浓度浸泡金莲花花蕾60 min为最优,诱导率为92.16%,且用时最短,是金莲花花药愈伤组织诱导的最佳预处理方式。本研究为今后的金莲花单倍体育种工作提供帮助,为金莲花遗传学研究提供基本材料。Taking Zhangjiakou traditional Chinese medicine Trollius chinensis as the test material, the effects of pretreatment with different mannitol concentrations and sucrose concentrations and 4℃ for different time on callus formation from anthers of Trollius chinensis were studied. The results showed that the effect of low temperature treatment at 4℃ was the best in 2 d. The induction rate was 85%, significantly higher than the non-low temperature pretreatment. Mannitol treatment was optimal for 3 days when the concentration was 0.3 mol/L. The induction rate reached 80.25%, significantly higher than other concentrations of mannitol treatment and control. Sucrose treatment with 0.1 mol/L concentration for soaking Trollius chinensis flower buds for 60 minutes was the best, and the induction rate was 92.16%. Significantly higher than other concentrations of treatment and control. The results showed that different pretreatment methods promoted the formation of Trollius chinensis callus, but soaking Trollius chinensis bud at 0.1 mol/L concentration for 60 min was the best, the induction rate was 92.16%, and the time was the shortest, which was the best pretreatment method for the induction of Trollius chinensis anther callus. This study could be helpful for the future work of Trollius chinensis haploid breeding and provides basic materials for the study of Trollius chinensis Bunge genetics.
