人参多糖对乙醇依赖小鼠戒断行为及血浆ACTH和皮质醇的影响

摘　　要：目的探究人参多糖(Ginseng polysaccharides,GSP)对乙醇依赖小鼠戒断行为及血浆促肾上腺皮质激素(Adrenocorticotropic hormone,ACTH)和皮质醇水平的影响。方法将昆明种小鼠100只,随机分为5组,空白组腹腔注射生理盐水,模型组和GSP 3个剂量组(100、200、400 mg/kg)腹腔注射2.5 g/kg乙醇(20%,v/v),连续15 d,并于最后一次给予乙醇后24 h进行明暗箱实验、高架十字迷宫实验和乙醇戒断症状评分。采用酶联免疫吸附法(ELISA)检测血浆中ACTH和皮质醇含量。结果中、高剂量GSP显著提高乙醇依赖戒断小鼠在明暗室中穿梭次数和在明室内停留时间以及进入开臂次数比和在开臂停留时间比。此外,GSP中、高剂量组小鼠戒断症状评分低于模型组。ELISA结果显示,GSP能显著降低血浆皮质醇含量,增加ACTH含量。结论GSP可能通过调节ACTH和皮质醇的含量改善乙醇戒断引起的焦虑相关行为。Objective To study the effects of ginseng polysaccharides(GSP)on withdrawal behavior and plasma ACTH and cortisol in ethanol-dependent mice.Methods Totally 100 Kunming mice were randomly divided into 5 groups.Control group was intraperitoneally injected with saline.Model group and GSP groups(GSP 100,200 and 400 mg/kg)were intraperitoneally injected with 2.5 g/kg ethanol(20%,v/v)for 15 d.The light-dark box experiment,the elevated plus maze test,and the withdrawal symptom score were performed at 24 h after the last administration of ethanol.Plasma ACTH and cortisol levels were measured by enzyme-linked immunosorbent assay(ELISA).Results GSP(200 and 400 mg/kg)significantly increased the number of cross and the ratio of the residence time in the bright room in the light-dark box experiment.The numbers of entries and time spent in the open arms were increased in GSP(200 and 400 mg/kg)groups.In addition,the scores of withdrawal syndrome in GSP(200 and 400 mg/kg)groups were lower than that in model group.ELISA results showed that GSP significantly decreased plasma cortisol level and increased ACTH content.Conclusion GSP improves the anxiety-related behavior induced by ethanol withdrawal by modulating plasma ACTH and cortisol levels.
