摘　　要：采用流式Annexin V-FITC/PI双染法检测对照组、DDP(顺铂)处理组、Mdivi-1+DDP处理组三组L1210细胞凋亡情况,用荧光探针DCFH-DA检测各组细胞内ROS水平,探讨Mdivi-1对顺铂诱导L1210细胞活性氧产生及细胞凋亡的影响。结果表明与对照组相比,DDP处理组L1210细胞凋亡数量明显增加,且细胞内ROS水平明显升高;Mdivi-1+DDP处理组细胞凋亡数量及ROS产生较DDP处理组均明显减少。Mdivi-1可抑制顺铂诱导的L1210细胞ROS产生及细胞凋亡。Flow Annexin V-FITC/PI double staining was used to detect the apoptosis of L1210 cells in the control group,DDP(cisplatin)treated group and Mdivi-1+DDP treated group.The level of ROS in each group was detected by DCFH-DA fluorescence probe to explore the effect of Mdivi-1 on the production of active oxygen and apoptosis of L1210 cells induced by cisplatin.The results showed that compared with the control group,the number of apoptosis and ROS level of L1210 cells in DDP treated group were significantly increased,and the number of apoptosis and ROS production in Mdivi-1+DDP treated group were significantly reduced compared with DDP treated group.Mdivi-1 can inhibit the Ros production and apoptosis of L1210 cells induced by cisplatin.
