摘 要:细胞焦亡是一种新发现的依赖于Gasdermin蛋白家族成孔活性的细胞程序性炎症坏死,在抵抗外部病原体入侵和感知内源危险信号中发挥着不可替代的作用,其形态学特征、发生及作用机制与细胞凋亡等典型细胞死亡方式有显著不同。焦亡由炎症小体激活Caspase-1的经典细胞焦亡途径和胞质LPS激活Caspase-4/5/11的非经典细胞焦亡途径而诱发。本文就近年来细胞焦亡的发现和命名、形态学和分子特征、分子机制和焦亡相关的疾病进行综述,为疾病预防诊断及临床药物的靶向治疗提供理论依据和现实的可行性。Pyroptosis is a newly discovered programmed inflammatory necrosis that depends on the pore-forming activity of the gasdermin protein family.Pyroptosis plays a key role in detecting and responding to intracellular pathogens.The morphological and molecular determinants of pyroptosis are significantly different from other patterns of characteristic cell death such as apoptosis.The activation pathways of pyroptosis are divided into the canonical pyroptosis pathway in which inflammatory activate caspase-1 and the noncanonical pyroptosis pathway in which cytosolic LPS activates caspase-4/5/11.In this paper,the discovery and naming,morphological and molecular characteristics,molecular mechanisms of pyroptosis and diseases associated with pyroptosis in recent years are reviewed,which provides theoretical basis and realistic feasibility for disease prevention and targeted therapy of clinical drugs.
