摘 要:目的探讨造血细胞激酶(Hck)基因对肺腺癌A-549细胞增殖和侵袭的影响及机制。方法利用干扰和过表达Hck质粒转染A-549细胞,用MTT法检测细胞增殖能力,Transwell小室检测细胞侵袭能力,细胞划痕试验检测细胞迁移能力,RT-PCR法检测Hck和上皮生长因子受体(EGFR)mRNA表达水平,Western blot法检测Hck和EGFR蛋白表达水平。RT-PCR法检测18例临床肺腺癌组织和癌旁组织中Hck和EGFR mRNA表达水平,Pearson法分析Hck mRNA和EGFR mRNA的相关性。结果过表达Hck后A-549细胞增殖能力、侵袭能力和迁移能力均升高(P <0.05),EGFR mRNA和蛋白表达水平也有所升高(P <0.05);干扰Hck表达后A-549细胞增殖能力、侵袭能力和迁移能力均下降(P <0.05),EGFR mRNA和蛋白表达水平也对应降低(P <0.05)。肺腺癌组织Hck和EGFR mRNA的相对表达量均高于癌旁组织。Pearson相关性分析结果显示,癌组织中Hck mRNA与EGFR mRNA的表达呈正相关(r=0.555,P=0.017)。结论本研究发现Hck可能通过改变EGFR的表达水平调节肺腺癌细胞的生物学功能。Objective To investigate the effect and mechanism of hematopoietic kinase(Hck) gene on proliferation and invasion of lung adenocarcinoma A-549 cells. Methods The A-549 cells were transfected with interfering and over-expressing Hck plasmid. The cell proliferation ability was detected by MTT assay, the invasiveness was detected by Transwell chamber, the cell migration ability was detected by cell scratch test, and the ability of cell migration was detected by cell scratch test. Hck and epithelial growth factor receptor(EGFR) mRNA expression levels were detected by RT-PCR, Western blot method Hck and EGFR protein expressions were detected by, Western blot and Hck and EGFR mRNA expressions were detected by RT-PCR in 18 cases of lung adenocarcinoma and adjacent tissues. The correlation between Hck mRNA and EGFR mRNA was analyzed by Pearson method. Results After overexpression of Hck, the proliferation, invasion and migration ability were increased(P < 0.05), EGFR m RNA and protein expression was also increased(P < 0.05), and the ability of cell proliferation, invasion and migration were also increased(P < 0.05). After interfering with the expression of Hck, the proliferation ability, invasion ability and migration ability were all decreased(P < 0.05), and the expression level of, EGFR mRNA and protein also decreased(P < 0.05). The relative expression levels of Hck and EGFR mRNA in lung adenocarcinoma tissues were higher than those in adjacent tissues. The results of Pearson correlation analysis showed that there was a positive correlation between the expression of Hck m RNA and EGFR mRNA in lung adenocarcinoma tissues(r = 0.555, P = 0.017). Conclusion Hck may regulate the biological function of lung adenocarcinoma cells by changing the expression level of EGFR.
