土贝母皂苷与拉米夫定联合用药体外抗HBV的药效学作用研究

摘　　要：目的以乙型肝炎病毒转染的人肝癌细胞株HepG22.2.15细胞为模型,观察土贝母皂苷与拉米夫定联合用药后对细胞的毒性、细胞分泌的乙型肝炎表面抗原(HBsAg)、乙型肝炎e抗原(HBeAg)及上清液中乙型肝炎病毒(HBV)DNA的复制的影响。方法将不同浓度组的土贝母皂苷与拉米夫定50.0μg/mL联合用药2、4、8 d后,通过四甲基偶氮唑盐(MTT)比色法检测联合用药对细胞的毒性,采用酶联免疫吸附试验(ELISA)检测联合用药后对上清液中HBsAg、HBeAg的作用,最后采用荧光定量聚合酶链反应(PCR)法检测联合用药对上清液中HBV DNA的影响。结果(1)联合用药对细胞无明显毒性。(2)联合用药2 d对HBsAg具有协同抑制作用,联合用药8 d仅土贝母皂苷2.0、16.0μg/mL 2个浓度组对HBeAg产生协同抑制作用。(3)荧光定量PCR检测发现,Q值越大,对HBV DNA的抑制作用越强。结论土贝母皂苷与拉米夫定联合用药对HBsAg、HBeAg及HBV DNA具有一定的协同抑制作用。Objective To observe the effects of tubeimoside combined with lamivudine on the cytotoxicity,HBsAg and HBeAg secreted by cells and HBV DNA duplication in the supernatant with human hepatocellular carcinoma(HCC)cell line HepG 22.2.15 transfected with hepatitis B virus(HBV)as the model.Methods Different concentrations of tubeimoside combined with 50.0μg/mL lamivudine were used for 2,4,8 d.The effect of combined medication on cytotoxicity was detected by MTT colorimetry.Its effects on HBsAg and HBeAg in the supernatant were tested by ELISA after the combination medication.Finally,the fluorescent quantitative PCR was used to detect the effect of combined medication on HBV-DNA in the supernatant.Results(1)The combined medication had no obvious toxicity to the cells.(2)The combined medication for 2 d had a synergistic inhibition effect on HBsAg,while the combined medication for 8 d only produced the synergistic inhibition effect on HBeAg in the two concentrations groups of tubeimoside 2.0μg/mL and 16.0μg/mL.(3)The fluorescence quantitative PCR detection revealed that the higher the Q value,the stronger the inhibition on HBV DNA.Conclusion Lamivudine combined with tubeimoside possesses a certain synergistic inhibition to HBsAg,HBeAg and HBV DNA.
