摘  要:目的通过高效液相色谱法(HPLC)测定不同产地甘草药材中甘草酸含量,以期为甘草药材质量评定提供依据。方法采用Agilent1260高效液相色谱仪,以乙腈为流动相A,以0.05%磷酸溶液为流动相B,进行梯度洗脱,检测波长为237nm,流速:0.8mL·min-1,柱温:30℃。结果甘草酸在0.0634~0.3172mg·mL-1浓度范围内与峰面积呈良好的线性关系,r2=0.9994,平均加样回收率为99.4%,RSD为0.60%,所建方法快速、简便、重现性好。结论3个省份11个产地甘草药材中甘草酸的含量差异较大,其中宁夏同心所产甘草含量最高,从各地平均值看,宁夏回族自治区>甘肃省>内蒙古自治区,且宁夏回族自治区与其他2个省份之间差异显著。省内间的差异显著结果表明,宁夏回族自治区产地之间差异较大,甘肃省和内蒙古自治区产地之间差异较小。OBJECTIVE The content of glycyrrhizic acid in different origins of licorice was determined by high performance liquid chromatography(HPLC),in order to provide a basis for the quality evaluation of licorice.METHODS Using Agilent 1260 high performance liquid chromatography,acetonitrile was used as mobile phase A,and 0.05%phosphoric acid solution was used as mobile phase B for gradient elution.The detection wavelength was 237 nm,the flow rate was 0.8 mL·min-1,and the column temperature was 30℃.RESULTS Glycyrrhizin showed a good linear relationship with the peak area in the concentration range of 0.0634-0.3172 mg·mL-1,r2=0.9994,the average recovery rate was 99.4%,and the RSD was 0.60%.The method was fast,simple and reproducible.CONCLUSION The content of glycyrrhizic acid in licorice of 11 producing areas in 3 provinces was quite different.The licorice content of Ningxia Tongxin was the highest.From the average of the localities,Ningxia Hui Autonomous Region>Gansu Province>Inner Mongolia Autonomous Region,and the differences between Ningxia Hui Autonomous Region and the other two provinces are significant.Significant differences between the provinces indicate that there is a large difference between the producing areas of Ningxia Hui Autonomous Region,and the difference between the producing areas of Gansu Province and Inner Mongolia Autonomous Region is small.
