梯度洗脱HPLC法同时测定复方阿昔洛韦乳膏中双组分的含量

摘　　要：目的建立梯度洗脱HPLC法同时测定复方阿昔洛韦乳膏中阿昔洛韦和维A酸的含量。方法采用色谱柱CNW Athena C18-WP(100?,4.6mm×250mm,5μm),以甲醇-水-2%冰醋酸溶液为流动相,梯度洗脱,流速:1.0mL·min-1,检测波长290nm,柱温:45℃,进样量:10μL。结果阿昔洛韦在21~210μg·mL-1范围内与峰面积呈良好的线性关系(r=1.0000,n=6),平均回收率为100.92%,RSD为1.11%(n=9),维A酸在0.7~7μg·mL-1范围内与峰面积呈良好的线性关系(r=0.9999,n=6),平均回收率为99.20%,RSD为0.86%(n=9)。结论本方法简便、可靠,可作为复方阿昔洛韦乳膏的含量测定方法。OBJECTIVE To develop an HPLC method to simultaneously determine the content of aciclovir and tretinoin in compound aciclovir cream.METHODS The assay was carried out on CNW Athena C18-WP(100?,4.6 mm×250 mm,5μm)column by HPLC gradient elution wih methanol water 2%glacial acetic solution used as mobile phase at a flow rate of 1.0 mL·min-1.The detection wavelength was 290 nm.The column temperature was 45℃.The injection volume was 10μL.RESULTS Aciclovir had a good linear relationship within the range of 21-210μg·mL-1(r=1.0000,n=6),the average recovery was 100.92%and the corresponding RSD was 1.11%(n=9).Tretinoin had a good linear relationship within the range of 0.7-7μg·mL-1(r=0.9999,n=6),the average recovery was 99.20%and the corresponding RSD was 0.86%(n=9).CONCLUSION This method is convenient and reliable,it can be used for the content determination of compound aciclovir cream.
