摘  要:目的初步研究大鼠坐骨神经损伤后华勒氏变性的相关分子信号传导通路。方法制备大鼠坐骨神经损伤模型,分别于术后12 h、24 h、3 d、7 d、14 d截取神经损伤远端,以免疫组化方法染色观察雪旺细胞Bungner带的形成,以及Westernblot测定各时间点ERK1/2和AKT蛋白表达。应用相应蛋白通路抑制剂后观察Bungner带形成及ERK1/2和AKT蛋白表达,初步探索华勒氏变性的相关分子信号传导通路。结果 Bungner带形成伴随着p-ERK表达增加。单纯阻断ERK磷酸化对Bungner带形成无明显影响,并伴随p-AKT表达增加。联合阻断ERK及AKT磷酸化可明显降低Bungner带形成数量。结论大鼠坐骨神经损伤后华勒氏变性Bungner带的形成与ERK及AKT通路联合调控相关。Objective To preliminarily explore the related molecular signal of Wallerian degeneration after sciatic nerve injury in rats. Methods The model of sciatic nerve injury in rats was prepared, and then the distal end of injuried nerve was sampled at 12 h, 24 h, 3 d, 7 d and 14 d after the surgery. The formation of Bungner band in schwann cells was observed by immunohistochemical staining and the protein expressions of ERK1/2 and AKT at each time point were determined by Western blot. After the application of corresponding protein pathway inhibitors, the formation of Bungner band and the expression of ERK1/2 and AKT proteins were observed preliminarily to explore the related molecular signals about Wallerian degeneration. Results The formation of Bungner band was coupled with elevated p-ERK1/2. Simply inhibit ting the phosphorylation of ERK had no significant effect on Bungner band formation but activated the AKT pathway. Combined inhibition of ERK and AKT can reduced the number of Bungner bands significantly. Conclusion The formation of Bungner band after sciatic nerve injury in rats is associated with the combined regulation of ERK and AKT pathways.
