摘  要:目的探讨人参皂苷Rb1对百草枯所致呼吸衰竭大鼠肺功能的影响及作用机制。方法将50只SD大鼠随机分为对照组、模型组、人参皂苷Rb1低、中、高剂量组,每组10只。除对照组,其余各组大鼠均用20%的百草枯溶液以40mg/kg的剂量灌胃,构建大鼠百草枯中毒呼吸衰竭模型,造模1天后各组给药治疗7天,对照组与模型组皮下注射等体积的0.9%NaCl溶液;给药期间每天检测大鼠体重变化,观察大鼠体征。给药结束后,用AniRes2005动物肺功能仪检测各组大鼠肺功能;ELISA法检测血清TNF-α、IL-6、IL-1β水平,肺组织用于计算肺湿干重比、HE染色观察病理变化,Western blot法检测GRP78、β-catenin、MMP2的表达水平。结果与对照组比较,模型组因百草枯导致大鼠呼吸衰竭严重,大鼠体重逐渐下降,肺功能下降显著(P=0.000),血清TNF-α、IL-6、IL-1β含量显著上升(P=0.000);与模型组比较,人参皂苷Rb1低、中、高剂量组肺功能显著提高(P<0.05);肺组织湿干重比均低于模型组(P=0.000),且大鼠血清TNF-α、IL-6、IL-1β含量均有所降低(P=0.000);HE染色结果显示,给药组大鼠肺组织肺损伤程度均得到改善;且各给药组GRP78、MMP2、β-catenin蛋白表达水平均显著下调(P<0.01)。结论人参皂苷Rb1对百草枯所致大鼠呼吸衰竭有显著治疗作用,其潜在机制可能是缓解炎症,降低肺组织内质网应激反应,以及抑制Wnt信号通路,减少MMP2的表达。Objective To investigate the effect and mechanism of ginsenoside Rb1 on lung function in rats with respiratory failure induced by paraquat.Methods Fifty SD rats were randomly divided into control group,model group and ginsenoside Rb1 low,medium and high dose groups,with 10 rats in each group.Except the control group,the rats in the other groups were treated with 20%paraquat solution at a dose of 40mg/kg to construct a model of paraquat poisoning respiratory failure in rats.After 1 day of modeling,each group was treated for 7 days.An equal volume of physiological saline was injected subcutaneously with the control group and model group.Changes in body weight of the rats were measured daily during the administration period,and the signs of the rats were observed.At the end of the administration,we used AniRes 2005 animal lungs function analysis to detect lung function of rats.Serum TNF-α,IL-6,IL-1βlevels,lung tissue detected by ELISA was used to calculate lung wet-to-dry weight ratio.HE staining was used to observe pathological changes.Western blot was used to detect GRP78,β-catenin,MMP2 expression Level.Results Compared with the control group,paraquat caused a severe respiratory failure in rats,and the body weight of the rats decreased gradually,lung function declined significantly(P=0.000).The levels of serum TNF-α,IL-6 and IL-1βincreased significantly(P=0.000).Compared with the model group,lung function increased significantly(P<0.05).The wet-to-dry weight ratio of lung tissue in the low,medium and high dose groups of saponin Rb1 was lower than that in the model group(P=0.000),and the serum levels of TNF-α,IL-6 and IL-1 were decreased in the rats(P=0.000).HE staining results showed that the lung injury degree of the lung tissue of the rats in the administration group was improved,and the expression levels of GRP78,MMP2 andβ-catenin in each administration group were significantly decreased(P<0.01).Conclusion Ginsenoside Rb1 has significant therapeutic effects on respiratory failure induced by paraquat.The
