摘 要:目的研究白芍总苷对缺血性脑损伤大鼠海马线粒体三磷酸腺苷(ATP)酶活性、ATP含量的影响及其海马组织保护作用。方法采用阻断大脑右侧中动脉的方法复制缺血性脑损伤大鼠模型。治疗组分别1次/天灌胃给予白芍总苷50、100、200mg/kg,假手术组和模型组同步给予0.9%NaCl注射液,疗程28天。行盲法神经功能评分,2,3,5-氯化三苯基四氮唑(TTC)染色法观察并计算脑梗死体积,干湿比重法计算脑组织含水量,HE法进行中枢海马病理学检查,透射电子显微镜(TEM)观察海马区线粒体超微结构变化;测定线粒体Na+-K+-ATP酶、Ca^2+-ATP酶、Mg^2+-ATP酶活性以及ATP含量,原子化学发光法测定线粒体游离Ca^2+浓度。结果与模型组比较,白芍总苷100、200mg/kg组能够明显降低缺血性脑损伤大鼠神经功能损伤评分(P<0.01),降低脑梗死体积(P<0.05或P<0.01),降低脑组织含水量(P<0.05),明显改善中枢海马区组织细胞病理性改变,抑制海马区线粒体超微结构病变;改善线粒体Na^+-K^+-ATP酶、Ca^2+-ATP酶、Mg^2+-ATP酶活性并提高ATP含量(P<0.05或P<0.01),降低线粒体游离Ca^2+浓度(P<0.05)。结论白芍总苷可能通过改善线粒体ATP酶活性、提高ATP含量、降低线粒体游离Ca^2+浓度、抑制线粒体损伤而对脑组织缺血性损伤起到一定的保护作用。Objective To study the effects of total glucosides of paeony on mitochondrial adenosine triphosphate(ATP)content and ATPase activity in hippocampus of the rats with ischemic brain injury and its protective effect on hippocampus.Methods The rat model of ischemic brain injury was established by blocking the right middle cerebral artery.The therapy groups were given intraperitoneal administration 50,100,200mg/kg once a day for 28 days.The rats in sham operation group and model group were given 0.9 sodium chloride solution simultaneously.Row blind neurological function score.The water content of brain tissue was calculated by dry-wet specific gravity method.The volume of cerebral infarction was calculated by TTC staining method.The pathological examination of hippocampal area by HE staining.The ultrastructural changes of mitochondria by electron microscope(TEM)was observation.The content of ATP and the activity of Na^+-K^+-ATPase,Ca^2+-ATPase,Mg^2+-ATPase were measured by colorimetric method.The content of free Ca^2+was measured by atomic chemiluminescence.Results Compared with model group group,the total glucosides of paeony 100,200mg/kg group could significantly reduce brain water content in rats with focal cerebral ischemia(P<0.05),reduce cerebral infarction volume(P<0.05 or P<0.01),significantly improve histopathological changes in hippocampal area,significantly improve the ultrastructural pathological changes of ultrastructural in hippocampal area;significantly improve the activity of Na^+-K^+-ATPase,Ca^2+-ATPase,Mg^2+-ATPase and the content of ATP(P<0.05 or P<0.01),reduce mitochondrial free Ca^2+concentration(P<0.05).Conclusion The total glucosides of paeony may protect cerebral ischemic injury by improving mitochondrial ATPase activity,increasing ATP content,reduce mitochondrial free Ca^2+concentration and inhibiting mitochondrial damage.
