摘  要:目的比较苯并[a]芘(BaP)恶性转化细胞T-16HBE-C1与非转化细胞16HBE全基因组表达谱差异,为进一步阐明BaP致癌机制和筛选BaP致癌生物学标志提供线索。方法选择BaP恶性转化细胞T-16HBE-C1与非转化细胞16HBE作为BaP致癌机制研究模型,利用人全基因组表达谱芯片Human mRNA OneArray?HOA v7 Microarrays筛选细胞间差异表达基因。继而对差异表达基因进行生物信息学分析,包括利用DAVID数据库对差异表达基因进行GO分析和KEGG通路分析。结果人全基因组表达谱芯片共发现689个基因在T-16HBE-C1和16HBE细胞中表达差异在2倍以上(P<0.05),其中339个基因在T-16HBE-C1细胞中的表达高于16HBE细胞,350个基因在T-16HBE-C1细胞中的表达低于16HBE细胞。差异表达基因涉及多种生物学改变,主要包括肿瘤信号转导、细胞增殖与凋亡、细胞代谢和细胞黏附等。结论人支气管上皮16HBE细胞经BaP恶性转化后基因表达谱发生明显改变,差异表达基因可能在BaP致癌中发挥作用,并可能作为BaP致癌的潜在生物学标志。Objective To further explore the mechanism of benzo[a]pyrene(BaP)-induced carcinogenesis and screen relevant potential biomarkers through comparing gene expression profile changes in BaP-transformed 16 HBE cells.Methods BaP-transformed human bronchial epithelial 16 HBE cells(T-16 HBE-C1)and non-transformed 16 HBE cells were selected as cell models of BaP-induced carcinogenesis.Human mRNA OneArray?HOA v7 Microarrays were used to analyze and compare gene expression profiles of 16 HBE and T-16 HBE-C1 cells.Then biological information,including Gene Oncology and KEGG pathways,was gathered from DAVID database.Results 689 genes were identified as differentially expressed genes via gene expression profile analysis between 16 HBE and T-16 HBE-C1 cells,among which 339 genes were up-regulated in T-16 HBE-C1 cells and 350 genes were down-regulated in T-16 HBE-C1 cells.Differentially expressed genes were involved in many biological functions,such as tumor signal transduction,cell proliferation and apoptosis,cell metabolism and cell adhesion.Conclusion Gene expression profile of BaP-transformed cell T-16 HBE-C1 had significantly changed.Differentially expressed genes might play important roles and act as potential biomarkers in BaP-induced carcinogenesis.
