细辛脑注射液被动皮肤过敏试验及对血清IgE的影响

摘　　要：目的研究细辛脑注射液在豚鼠被动皮肤过敏试验中是否有过敏反应,并观察细辛脑注射液对血清IgE含量的影响。方法将豚鼠随机分为4组:阴性对照组、细辛脑注射液低、高剂量组和阳性药物组,分别腹腔注射生理盐水、细辛脑注射液和阳性对照物致敏,分离致敏豚鼠血清,采用Elisa法测定IgE含量;将血清稀释1∶2和1∶8皮内注射于正常豚鼠被动致敏进行被动皮肤过敏试验,测量背部蓝斑直径,统计过敏反应率。结果当抗血清稀释比例为1∶2和1∶8时,除阴性对照组未出现蓝斑,细辛脑注射液低、高剂量组和阳性对照组豚鼠背部均出现明显蓝斑,出现阳性反应;细辛脑注射液高剂量组和阳性药物组血清中IgE与阴性对照组相比均有显著性升高(P<0.05,P<0.01)。结论细辛脑注射液可诱发豚鼠皮肤被动过敏阳性反应。Objective To study whether asarone injection has allergic reaction in guinea pig passive cutaneous anaphylaxis(PCA)test,and its influence on serum IgE content.Methods Guinea pigs were randomly divided into the negative control group,low dose asarone injection group,high dose asarone injection group and positive drug group.Animals in each group were sensitized by intraperitoneal injection of saline,asarone injection or positive control.The serum of sensitized guinea pigs was collected to detect the concentration of IgE by ELISA.PCA were operated on normal guinea pigs by intradermal injection with the sensitized serum.Due to effusions of Evans blue,blue blot appeared on guinea pigs′back.The diameter of the blue blot was measured to assess the degree of allergic response.Results The negative control group showed no allergic reaction,as no blue blot was detected.The obvious blue spot on the back of the guinea pig was observed in the low or high dose asarone injection group and the positive control group.The IgE concentration was significantly higher in high dose asarone injection group and positive drug group than that of negative control group(P<0.05,P<0.01).Conclusion The asarone injection may cause the passive cutaneous anaphylaxis reaction in guinea pig.
