芸豆凝集素及壬苯醇醚对蚯蚓内外表皮作用的比较

摘　　要：[目的]探究芸豆凝集素(KBL)凝胶剂及常用避孕杀精剂壬苯醇醚(Nonoxynol-9,N-9)对蚯蚓(爱胜蚓属,Eisenia)内外表皮细胞的作用及损伤程度。[方法]分别用自来水将KBL凝胶制剂和N-9稀释为浓度分别为0.05、0.1、0.2、0.5、1、2 mg/m L的溶液,对照组为自来水。将蚯蚓放入各组溶液中浸泡培养,并用台盼蓝进行活体染色,在不同时间肉眼观察蚯蚓的存活情况,同时取材进行Bouin固定并制作石蜡切片,显微镜下观察不同实验组体表上皮细胞和消化道上皮细胞损伤程度。[结果]相同浓度的N-9与KBL凝胶剂,N-9对上皮细胞的刺激损伤远大于KBL;浓度≤2 mg/m L及以下的KBL凝胶制剂不会对蚯蚓上皮细胞造成损伤,反之N-9溶液在≥0.5 mg/m L时即可破坏蚯蚓上皮细胞,且N-9浓度越高,对蚯蚓的损伤程度越明显。[结论]浓度≤2 mg/m L及以下的芸豆凝集素凝胶剂不会对内外上皮细胞造成损伤,可以有效用于避孕,其抗病毒作用有助于减少性传播疾病的风险。[Objective]To explore the difference between kidney bean lectin and Nonoxynol-9 when acting on skin and intestinal epithelium of Eisenia.[Method]Diluting the KBL gel and Nonoxynol-9 with tap water to concentration of 0.05,0.1,0.2,0.5,1,2 mg/mL respectively,the control group is tap water.Earthworm was immersed in KBL or N-9 solution and stained by Trypan blue solution,then observed through naked eye and microscopic examination by making transverse section after fixed and packed in wax to conclude whether the skin and intestinal epithelium of Eisenia was damaged.[Result]The damage was more obvious in N-9 than KBL solution when cultured in the same concentration;When the concentration of KBL gel was 2 mg/mL or below,the skin and intestinal epithelium didn’t be damaged;In contrast,when the concentration of N-9 was 0.5 mg/mL or above,the skin and intestinal epithelium of earthworm was damaged badly and the higher concentration,the worse damage.[Conclusion]KBL gel didn’t damage the skin and intestinal epithelium of earthworm,and can be used for both contraception and may be used to prevent the sexual transmitted diseases including HIV.
