摘  要:目的探讨妊娠期糖尿病(GDM)患者外周血胎盘来源外泌体中miRNA的表达情况,评估其在GDM早期预测中的价值。方法选取2018年1月至12月在南方医科大学附属东莞人民医院诊治的GDM患者25例、健康孕妇25例。利用密度梯度离心法分离血清中胎盘来源外泌体;通过基因芯片和实时荧光定量PCR检测miRNA表达情况;建立logistic回归模型,以ROC曲线分析其预测效能。结果GDM患者胎盘来源外泌体中存在7种异常表达的miRNAs,其中miR-122、miR-29a、miR-210、miR-20、miR-192表达显著上调,而miR-223、miR-130a表达明显下调。7种miRNA联合检测AUC值为0.903(95%CI 0.813~0.992)。结论胎盘来源外泌体中异常表达的miRNA可作为GDM早期预测的潜在标志物,值得深入研究。ObjectiveTo investigate the miRNA expression of placenta-derived exosomes in patients with gestationaldiabetes mellitus(GDM),and evaluate its value in early prediction of GDM.MethodsA total of 25 pregnant womenwith GDM and 25 healthy controls were recruited from January 2018 to December 2018 at Dongguan People’s Hospital ofSouthern Medical University.The placenta-derived exosomes were extracted by density gradient centrifugation.Themi RNA expression was assessed using microarray and real-time quantitative PCR.Logistic regression model wasestablished and ROC curve was applied to evaluate its predictive value.ResultsA total of 7 dysregulated mi RNAs wereidentified,including 5 up-regulated mi RNAs(miR-122,mi R-29 a,miR-210,miR-20,miR-192)and 2 down-regulated mi RNAs(miR-223,miR-130 a).The AUC of 7-mi RNA panel was 0.903(95%CI 0.813~0.992).Conclusion Dysregulated mi RNAs of placenta-derived exosomes have potential value in early prediction of GDM,and deservefurther investigation.
