摘  要:目的观察针刺对卒中后抑郁(PSD)大鼠行为学改变及脑组织中脑源性神经营养因子(BDNF)、胶质纤维酸性蛋白(GFAP)表达水平的影响,探讨针刺治疗PSD的机制。方法从60只雄性SD大鼠中随机选取10只入正常组,其余造模。采用大脑中动脉线拴阻塞法复合慢性不可预知的温和应激法建立大鼠PSD模型,将造模成功的大鼠随机分为模型组、针刺组、氟西汀组。针刺组针刺百会(GV20)、风府(GV16)、神门(HT7)、太冲(LR3),每日1次,氟西汀组每日1次灌胃(2 mg/kg),均连续治疗3周。实验过程中观察各组大鼠的行为学改变,蛋白免疫印迹法检测海马、皮质、杏仁核BDNF、GFAP蛋白的表达情况;RT-PCR法检测海马、皮质、杏仁核BDNF、GFAP mRNA的表达情况;通过透射电镜观察脑组织内星形胶质细胞超微结构的改变。结果与正常组比较,模型组大鼠海马、皮质、杏仁核的BDNF、GFAP蛋白及BDNF、GFAP mRNA表达水平增加(P<0.01或P<0.05);与模型组相比,针刺组与氟西汀组大鼠海马、皮质、杏仁核中BDNF、GFAP蛋白及BDNF、GFAP mRNA表达水平增高(P<0.01)。结论针刺可以改善PSD大鼠的症状,其作用机制可能与针刺提高大鼠海马、皮质和杏仁核BDNF、GFAP的表达,增强星形胶质细胞ATP的释放,进而改善神经元损伤,促进神经元再生有关。Objective To observe the influence of acupuncture on behavioral changes and expressions of brain-derived neurotrophic factor(BDNF)and glial fibrillary acidic protein(GFAP)in hippocampus,cortex and amygdala in rats with post-stroke depression(PSD),and discuss the mechanism of acupuncture treating PSD.Methods Male SD rats(n=60)were randomly divided into normal group(n=10)and modeling group(n=50).The rat model was established by using thread approach of middle cerebral artery occlusion(MCAO)combined with chronic unpredictable mild stimulation(CUMS).The successful modeling rats were randomly divided into model group,acupuncture group and fluoxetine group.The acupuncture group was given needling in Baihui(GV20),Fengfu(GV16),Shenmen(HT7)and Taichong(LR3)once a day for 3 weeks,and fluoxetine group was orally given fluoxetine(2 mg/kg)once a day for 3 weeks.The behavioral changes were closely observed,and protein expressions of BDNF and GFAP in hippocampus,cortex and amygdale were detected by suing Western blotting assay in all groups.The mRNA expressions of BDNF and GFAP in hippocampus,cortex and amygdale were detected by using RT-qPCR,and ultrastructural changes of astrocytes(Ast)in brain tissue were observed by using transmission electron microscope(TEM).Results Compared with normal group,the protein and mRNA expressions of BDNF and GFAP in hippocampus,cortex and amygdale all increased in model group(P<0.01 or P<0.05).Compared with model group,the protein and mRNA expressions of BDNF and GFAP in hippocampus,cortex and amygdale all increased in acupuncture group and fluoxetine group(P<0.01).Conclusion Acupuncture can relieve the symptoms in PSD rats,and the mechanism may related to that it can promote the expressions of BDNF and GFAP in hippocampus,cortex and amygdale,improve ATP release in Ast,ameliorate neuron injury and improve neurogenesis.
