摘 要:目的:探究生精汤对少弱精子症小鼠精液质量,SOD、MDA、GSH-Px水平以及精子线粒体m PTP开放的影响,并深入研究其作用机制。方法:用雷公藤多苷给雄性小鼠连续灌胃四周建立模型,将其分为空白对照组、阳性对照组(益肾灵颗粒组)、模型组、生精汤高剂量组(60g/kg)、低剂量组(15g/kg),给药4周后,采用CASA技术检测精子活力、密度、总数等指标来评价精液质量,选取相应的试剂盒检测SOD、MDA、GSH-Px水平,荧光分光光度仪检测精子线粒体m PTP吸光度值。结果:模型组精子总活力、精子密度、SOD、GSH-Px水平、线粒体吸光度值较空白组显著降低,MDA水平显著升高,且有显著性差异(P<0.05),提示造模成功;经给药治疗后,生精汤高、低剂量组精子总活力、精子密度,SOD、GSH-Px水平、线粒体吸光度值较模型组显著升高(P<0.05),MDA水平显著降低(P<0.05)且趋于空白对照组。结论:生精汤对少弱精子症小鼠有一定的治疗作用,其作用机理可能与提升精子线粒体m PTP,降低精子氧化应激反应从而来增强精子活力及密度有关。Objective:To investigate the effect of Shengjing Decoction on semen quality,SOD,MDA,GSH-Px concentration and sperm mitochondrial m PTP opening in mice with oligozoospermia and asthenospemiia,and to study its mechanism.Methods:Male mice were continuously perfused with tripterygium glycosides to establish a mouse model,which was divided into blank controlled group,positive controlled group(Yishenling Granule Group),model group and Shengjing Decoction high dose group(60g/kg),low dose group(15g/kg),after 4weeks of administration,the quality of sperm,density,total number and other indicators were used to evaluate the quality of fsemen.The corresponding kit was used to detect SOD,MDA,GSH-Px concentration,luorescence spectrophotometer to detect sperm mitochondrial m PTP absorbance value.Results:The total sperm motility,sperm density,SOD,GSH-Px concentration,sperm mitochondrial absorbance value of the model group were significantly lower than those of the blank group,MDA concentration was significantly increased,and there was a significant difference(P<0.01),suggesting successful modeling.After administration,the total sperm motility,sperm density,SOD,GSH-Px concentration and sperm mitochondrial absorbance of Shengjing Decoction Group were significantly higher than those of the model group(P<0.01),and the MDA concentration was significantly decreased(PvO.01),and tended to the blank controlled group.Conclusion:Shengjing Decoction has certain therapeutic effects on mice with oligozoospermia and asthenospermia,and its mechanism may be related to improving sperm mitochondrial m PTP and reducing sperm oxidative stress to enhance sperm motility and density.
