摘 要:目的以捕获的未知鼠为对象,应用DNA条形码技术进行鼠种鉴定,验证该方法在啮齿动物鉴定的可行性。方法以动物线粒体细胞色素C氧化酶亚基I(COI)作为目标基因,PCR扩增、测序。测序结果在美国国立生物技术信息中心(NCBI)网站上运用基于局部比对算法的搜索工具(BLAST)程序进行序列同源性比较和分析,检验其结果是否与该动物的外部形态和头骨标本鉴定结果相同。结果样本线粒体COI基因与基因库(GenBank)中编号为KJ152220.1的社鼠(Niviventer confucianus)同源性为99.01%,再结合形态学鉴定结果,可以确定该未知鼠种为社鼠,DNA条形码鉴定结果与形态学鉴定结果相一致。结论DNA条形码技术可作为形态学鉴定的辅助方法,能用于鼠类标本的分类鉴定。Objective To verify the feasibility of DNA barcoding technique in rodents identification,we apply methods in an unknown mice captured in the monitoring.Methods Animal Mitochondrial cytochrome C oxidase subunit I(COI)gene was used as the target gene,then PCR amplification and DNA sequencing were performed.The sequencing results was used for sequence homology comparison and analysis on NCBI website by using BLAST program in order to test whether the results accord with the animal’s external shape and skull specimen identification finding.Results The homology of mitochondrial COI gene sequence of sample and the rat(GenBank:KJ152220.1)was 99.01%.Condusion with morphological identification results,this unknown rat could be identified as Niviventer confucianus.The results of DNA barcoding identification were consistent with those of morphological identification.Conclusion DNA barcode technology can be used as an auxiliary method in morphological identification and classification of the specimens of rats.
